The experiment will be conducted to research the specific impacts of lycopene β-cyclase interference on carotenoid metabolism, cherry tomato(
Solanum lycopersicum
L. var. c
erasiforme
Alef.) fruitspecific lycopene β-cyclase interference vector was constructed, and the validity in different color fruits were verified. Cherry tomato fruit-specific promoter E8 was amplified according to X13437.1 and fruit-specific vector of E8-pBI121 was constructed, which could be expressed in cherry tomato fruits of pink, red, green and purple colors. According to X86452.1,two different lycopene β-cyclase sequences
B
1 and
B
2 were amplified,
LYC-B
1 with 801 bp from 61 bp to 861 bp and
LYC-B
2 with 302 bp from 480 bp to 781 bp of
LYC-B
. The interference vector of pBI121-B1B2 with CaMV 35S promotor and the fruit-specific expression interference vectors of E8- pBI121- B1B2 with E8 promotor were constructed. E8-pBI121,pBI121-B1B2 and E8-pBI121-B1B2 were transferred into
Agrobacterium tumefaciens
GV3101 with the freeze-thaw method, then transformed into living cherry tomato fruits and leaves by injection. The results of GUS staining 7 days after injection showed that E8-pBI121 expressed in pink, red, green and purple cherry tomato fruits;pBI121-B1B2 expressed in leaves, fruits and seeds, while E8- pBI121-B1B2 expressed just in fruits and seeds.

Toxin-producing Condition Optimization and Biological Activity Determination of
Colletotrichum orbiculare
TANG Shuang-Shuang, LIU Zhi-Heng, YU Chao-Ge, ZHENG Chuan, LI Jian-Bing, ZHAO Ting-Chang
Toxin-producing condition optimization and determination of crude toxin biological activity and its physical, chemical properties were carried out on
Colletotrichum orbiculare
. The results showed that different culture conditions had different influences on pathogenic toxin activity. The Richard culture medium with pH 6 ,culture temperature at 25℃,in dark, and shaking continuously for 20 d were the best toxin-producing conditions. The watermelon stem inoculated by crude toxin could produce the same acute symptom as the bacteria did, which had inhibitory effect on seed radicle growth and seed germination; and also a collapsing role on seedling. That toxin is the main pathogenic factor in the process of watermelon anthrax bacteria infection. Among the 10 kinds of selected plants inoculated by crude toxin, the symptoms of muskmelon, summer squash and strawberry were significant, while no symptoms were seen on tomato, eggplant and pepper.

SRAP Analysis of Mustard Germplasm Genetic Diversity and Genetic Relationship
LI Ning, SI Jun, REN Xue-Song, SONG Hong-Yuan, LI Cheng-Qiong
Abstract:The paper analyzed the genetic diversity of 111 mustards〔
Brassica juncea
(L.)Czern. et Coss.〕 materials using SRAP markers, and recorded their morphological characteristics.21 primer combinations with high polymorphism were screened from 300 SRAP primer combinations.106 polymorphic bands among 150 bands were amplified by 21 SRAP primer combinations, and the polymorphic rate was 70.67%.The SRAP clustering results showed that the genetic similarity of 111 mustards ranged from 0.38 to 0.89,and 111 mustards were divided into 5 groups and several subgroups. The agronomic traits including leaf blade traits, and root, stem, leaf, bolting development were investigated and analyzed. The results showed that the genetic similarity of 111 mustards ranged from 0.39 to 1.00,and they could be divided into 4 groups and several subgroups. These two results were nearly the same, indicating that SRAP markers could be applied in studying the genetic diversity and relationships of mustard germplasm resources.

Effects of Saline Stress on Growth and Rhizoplane Soil Enzyme Activities of Cucumber Seedling Grafted on Different Rootstocks
ZHAO Yuan, WU Feng-Zhi
Based on the saline content and composition of the soda saline soil in Heilongjiang and other Provinces, this experiment simulated its saline composition, adopted pot culture, grafted cucumber(
Cucumis sativus
) seedlings onto 2 salt-tolerant rootstocks, and 2 salt-sensitive rootstocks as experimental materials. Ownroot seedlings were taken as the contrast. Saline solution(100 mmol·L
-1
,pH 9.0) was used to treat the soil. We studied the effects of salt stress on growth and soil enzyme activities of cucumber seedlings grafted on these different rootstocks. The results indicated that the yield per plant, the ground fresh weight, the root fresh weight, the plant height and the chlorophyll content of cucumber seedlings grafted on 2 salt-tolerant rootstocks were significantly higher than that of the 2 salt-sensitive rootstocks and own-root cucumber. The soil urease, neutral phosphatase activities after transplanted 30 d and the catalase, dehydrogenase activities after transplanted 40 d of cucumber seedlings grafted on salt-tolerant rootstocks were higher than that of salt-sensitive rootstocks and own-root plants. These 4 kinds of soil enzyme activities of cucumber seedlings grafted on salt-tolerant rootstocks after transplanted 50 d were all higher than that of salt-sensitive rootstocks and own-root cucumber. Under soil salinization condition, cucumber seedlings grafted on salt-tolerant rootstocks grew robustly and suffered low degree of salt injury. That account for that graft can significantly improve cucumber salt resistance; the resistance enhancement of cucumber seedlings grafted on salt-tolerant rootstocks is closely related to the increase of soil enzyme activity.

CHEN Qi-Wu, YANG Xiao-Yun, LUO Qing-Xi, ZANG Shun, ZHANG Shu-Xia, SI Chao-Guang, WANG Yuan, ZHANG Qing-Xia
Taking Chinese cabbage〔
Brassica campestris
L. ssp.
pekinensis
(Lour.)Olsson〕line 03B9 with high sensitivity and C24 with high resistance to black sesame spot as experimental materials, this paper studies the effect of different copper concentration treatments on black sesame spot, and total phenol content and PPO activity. The results showed that the sesame spots of the 2 lines were markedly increased as the copper concentration increased, and the occurrance degree of 03B9 was significantly higher than that of C24. During the copper treatments, the total phenol content,PPO activity in the petiole of line C24 were higher than that in line 03B9 with 25.6% and 77.0%,respectively. The preliminary tests showed that under hydroponic conditions, the occurrence of black sesame spot was promoted by the concentration of exogenous copper, the degree of disorder was related to the copper treatment concentration. Meanwhile, the total phenol content and PPO activity were influenced by copper.
