1 Effect of Plant Growth Substances on Sex Differentiation of Cucumber

This paper expounds the effect of plant growth substances on sex differentiation of cucumber(Cucumis sativus L.).The substances include ethylene,gibberellin,auxin,polyamines,and brassinolide.It also introduces the situation of researches about molecular mechanism of cucumber sex differentiation,and puts forward direction for future research.

14 Segregate Cloning and Sequence Analysis of lea3 Gene Complete cDNA in Zizania caduciflora

Group 3 later embryogenesis abundant proteins(LEA3),present in abundance in seeds during the late stages of development,induced by exogenous abscisic acid(ABA),and associated with drought tolerance.According to high conservative amino acids of reported LEA3 in Gramineae,degenerate primers were designed and a special DNA fragment was obtained from Zizania caduciflora by RT-PCR.Then based on the obtained sequence,some new primers were designed, and two terminals of lea3 gene were cloned by 3′RACE and 5′RACE respectively.Splicing the fragments of sequence,the full length cDNA sequence was obtained which indicated that open reading frame region of this gene was 666 bp,coded 221 amino acid,including 2 characteristic motif of lea3 gene only.Through comparing with the reported lea3 genes sequences,we found that Zizania caduciflora shared the identity with Oryza sativa,Hordeum vulgare,Triticum aestivum,Bromus inermis and Zea mays as 51.2 %,41.0 %,41.1 %,42.0 % and 40.5 %,respectively.It was concluded that Zizania caduciflora lea3 gene had genetic characteristics similar with all species and it could provide reference for functional research.At present,this nucleotide sequence data are deposited in GenBank databases under the accession number GQ494017.

19 Genetic Diversity Analysis of Fusarium oxysporum Isolated from Watermelon,Melon and Cucumber with UP-PCR

Twentyone isolates of Fusarium oxysporum from watermelon(Citrullus lanatus),melon(Cucumis melo L.)and cucumber(Cucumis sativus L.)in various areas of Liaoning province were analyzed by UP-PCR.The results indicated that a total of 61 bands were amplified using 6 primers,of which 58 amplified fragments(95.1 %)were polymorphic.The isolates were clustered into six distinct groups at the similarity of 0.746 by cluster analysis.FOG1 group includes six strains of melon,FOG2 group includes six strains of watermelon,FOG3 group includes six strains of cucumber,FOG4 includes a strain of watermelon,FOG5 includes a strain of melon,FOG6 contains a strain of cucumber.

24 Correlation and Principal Component Analyses on Yield Traits of Varieties with Different Ripe Period in Phaseolus vulgaris L.var.chinensis Hort.

In this paper,P.vulgaris L.var.chinensis Hort.was analyzed through statistical method by correlation and principal component analyses.The result shows that there are comparatively large differences between early maturing and late maturing varieties.The results of correlation analysis shows that there are 17 couples of traits have significant or very significant correlation value in early maturing varieties,9 couples of traits have significant or very significant correlation value in late maturing varieties,and only 4 couples of traits have significant correlation value among these 2 types of varieties.The result of principal component analysis shows that there are 4 principal components among early maturing and late maturing varieties,whose characteristic values exceed 1.This can be explained variance(r=83.612 %、r=84.293 %).In the first principal component,the trait of pod width,pod thickness,yield per plant,fresh pod weight and pod length have bigger weight coefficient in early maturing and late maturing varieties,which are major output factors of P.vulgaris L.var.chinensis Hort.The other 3 principal components have different contribution for agronomic traits.

28 Identification of Physiological Races of Powdery Mildew on Melon in Gansu Province

Ten pathogen collections of powdery mildew,came from different melon(Cucumis melo L.)grown areas of Gansu province,were detected by standard system for identification of powdery mildew.According to the internationally recognized melon differential standards for identifying races of powdery mildew,we preliminarily confirmed that there were race 1 and race 2France of Podosphaera xanthii,and additional physiological race 7 of powdery mildew in Gansu province.Race 1 of Podosphaera xanthii occurred more frequently,which was the prevailing race.

33 Evaluation of Tomato Resistance to Meloidogyne incognita

In order to study on the main northern 90 tomato(Lycopersicon esculentum Mill.)cultivars’resistance to Meloidogyne incognita,tomato plants were inoculated artificially to identify resistance index in solar greenhouse.The results revealed that in tested cultivars:‘Fentailang No.1’,‘Fentailang No.2’,‘HortPioneer’,0745,‘Jinguan No.1’and‘Baili No.2’showed high resistance,‘Jiahong No.6’,‘Xianke No.1’,‘06Chunzhan No.7’,‘Jiayuandafen’and‘Jinguan No.2’revealed moderate resistance,‘06Chunzhan No.8’,‘06Qiu 88’,‘Israel 405’,‘Jinguan No.5’and‘Jinguan No.7’showed resistance.The rest cultivars were susceptible or high susceptible to Meloidogyne incognita.For the tested cultivars in this research,there was no immune one.

38 Study of Premature Bolting of Carrot in Spring Cultivation

The premature bolting of carrot(Daucus carota L.var.Stiva DC.)in spring cultivation was investigated with 101 accessions including five ecotypes in 2004,2005 and 2008.The results showed that the premature bolting ratio of 37 accessions of Danvers,Nantes,Amsterdam and Kuroda were zero,and the ratio of one Nantes,one Kuroda and three purple accessions were more than 20 %.The average ratio of purple type was about 48.4 %,which was significantly higher than other ecotypes.The average ratio of 2004 was 7.0 %,which was significantly higher than that of 2005(2.7 %)and 2008(3.7 %).The premature bolting of carrot could be induced by long-day.

43 Studies on Leaf Area Indexes of Cabbage and Cauliflower Seedling

In this study,the leaf area indexes of 3 cabbage varieties,3 cauliflower varieties,2 crops Brassica oleracea L.var.capitata L.and Brasica oleracea L.var.botrytis L.were worked on and validated by the assay method about seedling leaf area.The seedling leaf area indexes of Brassica oleracea L.var.capitata L.and Brasica oleracea L.var.botrytis L.are 0.681 and 0.630,and these two leaf area indexes have significant deviation,so that these 2 crops can not share one pooling leaf area index.

47 Effects of pH Value of Culture Medium on Chinese Kale(Brassica alboglabra L.H.Bailey)in Vitro Regeneration

After autoclave sterilization,changes of different pH value of MS medium were compared.Hypocotyls and adventitious buds were cultured in culture medium with different pH value to discuss the effect of pH value on in vitro regeneration.The results showed that after autoclave sterilization,pH value of MS medium with different pH value declined in varying degrees.The most appropriate differentiation medium for hypocotyls of Chinese kale(Brassica alboglabra L.H.Bailey)was the treatment with 6.20 pH value before sterilization and 5.65 after sterilization.The most suitable rooting mediums were the two treatments with pH value higher than 6.00 after sterilization.

50 Preliminary Studies of Silicon Surfactant Ag-64 on the Chemical Control Effects of Spodoptera exigua in Green Chinese Onion

In order to explore the role of silicon surfactant on the control effect of beet armyworm(Spodoptera exigua),S.exigua Nuclear Polyhedrosis Virus(SeNPV),emamectin benzoate,and chlorantraniliprole were added silicon surfactant Ag-64(0.1 %)respectively,and 25 % effective dose,50 % spray volume were cut down for each insecticides in the meantime.Each treatment were sprayed at the peak of 1st-3rd instar larvae of Spodoptera exigua and the same or better control effects could be obtained,suggesting that the silicon surfactant could increase the moisture quality on the leaves of green Chinese onion(Allium fistulosum L.var.giganteum Makino).

53 Effects of Rhizosphere Inoculation with Bacillus licheniformis and Bacillus polymyxa on Pepper and Cucumber Seedling Development

Bacillus licheniformis and Bacillus polymyxa were inoculated by suspensions injection into pepper(Capsicum annuum L.)and cucumber(Cucumis sativus L.)seedling rhizosphere,and the seedling development and their relative physiological targets were analyzed.The results indicated that after rhizosphere inoculation in pepper seedling significantly increased net photosynthetic rate and root vigor,and the absorption of mineral elements such as N,P,K,Ca,Mg,Fe were enhanced.The pepper yield was increased by 29.3 % to 33.3 %.B.polymyxa showed better effect than B.licheniformis.Contrast with pepper,B.licheniformis and B.polymyxa inoculation in cucumber seedling rhizosphere decreased the root vigor,thus remarkably inhibited mineral elements absorption and fruit production.

58 Studies on Effects of Three Arbuscular Mycorrhizal Fungi Inoculation on Pepper Root System

In this experiment,as experimental material,‘Jinta’pepper(Capsicum annuum L.)was inoculated with three arbuscular mycorrhizal fungi(G.etunicatum,G.mosseae,G.versiforme)during sowing period.After mycorrhizal seedling planting,they were transplanted to field.Changes in pepper root growth and enzyme activity during the entire growth process were studied.The results showed that root biomass,root activity,root SOD,POD,CAT activities and yield of the 3 inoculation treatments were significantly higher than that of the control.Among all the 3 experimental strains,G.mosseae was the best for promoting pepper root growth.

63 Field Insecticides Trial against Cowpea Aphid(Aphis craccivora)and Liriomyza sativae

Insecticides of 2.5 % imidacloprid EC,2.5 % lambda-cyhalothrin EC,5 % acetamiprid EC,100 g·L-1 alpha-cypermethrin EC and 25 % buprofezin WC were used for field trial against cowpea aphid(Aphis craccivora).Insecticides of 480 g·L-1 chlorpyrifos EC,4.5 % cypermethrin EC,80 % dichlorvos EC,75 % cyromzinge WP and 0.5 % emamectin benzoate EC were used for field trial against Liriomyza sativae.The results showed that 1 500 times dilution of 2.5 % imidacloprid EC,2 000 times dilution of 2.5 % lambda-cyhalothrin EC,1 000 times dilution of 5 % acetamiprid EC and 2 000 times dilution of 100 g·L-1 alpha-cypermethrin EC could effectively control cowpea aphid and the efficacy were all above 95.76 % during 1 day to 10 days after control.The efficacy of 1 000 times dilution of 25 % buprofezin WC was very low and it was only 66.68 % in 3 days after spraying.3 000 times dilution of 75 % cyromzinge WP had good control effects on Liriomyza sativae and its efficacy was 89.63 %,94.10 % and 93.28 % differently after 3 days,7 days and 10 days.The secondary efficacy was given by 480 g·L-1 chlorpyrifos EC,with control effect of 61.12 %,69.15 % and 69.40 % after 3 days,7 days and 10 days.The other insecticides all had very low efficacy.

67 Effects of Foliar Zinc Application on Amaranth(Amaranthus mangostanus L.)Growth

0.28 ml·L-1,0.40 ml·L-1,1.00 ml·L-1 concentration of zinc fertilizer were sprayed on leaf surface of amaranth(Amaranthus mangostanus L.).The effects on amaranth growth,quality and yield were studied.The results showed that 3 zinc application treatments could increase amaranth height,stem diameter and leaves area,improve its quality,decrease the nitrate accumulation,and increase yield.Treatment with concentration of 0.40 ml·L-1 had the best effect.

70 Preliminary Studies on Effects of Solar-shading Screen Cover on Quality of Wettish-cultivated Water Dropwort〔Oenanthe stolonifera(Roxb)Wall.〕

Using cultivation with solar-shading screen cover,we studied the effect of solar-shading screen cover on the quality of wettish-cultivated water dropwort〔Oenanthe stolonifera(Roxb)Wall.〕.The results showed that the contents of VC,soluble protein,carotenoid,manitol,and total flavone of leaflets were significantly decreased,but the chlorophyll content was significantly increased.The contents of crude fiber of petiole were significantly lower than the contrast,but contents of the total dietary fiber was basically the same as the contrast,the contents of soluble dietary fiber was significantly higer than the contrast.

74 Correlation Analysis on Quantitative Traits of Tomato Germplasm Resources

Correlation analysis were conducted on 14 tomato(Lycopersicon esculentum Mill.)quantitative traits and lycopene content by using DPS software.The results showed that the lycopene content is very significantly positively correlated with single inflorescence flower numbers,single inflorescence fruit numbers and soluble solids content,but very significantly negatively correlated with size of cordy area around pedicel scar,pedicel length and single fruit weight.The lycopene content is significantly positively correlated with fruit shape index,but significantly negatively correlated with fruit firmness,flesh thickness,longitudinal diameter fruit.

77 Studies on Selection of Restoring Line of Pepper Cytoplasmic Male Sterile Line 8A

As the male parents,56 single plants out of seven inbred lines were crossed to pepper(Capsicum annuum L.)CMS line 8A,and the resulting F1 hybrids were used for the identification of restoration lines.The results showed as following:the fertile plants generated plenty of pollen grains which were dyed by TTC and took on red,while,the sterile plants generated no or few pollen grains which were dyed by TTC and took on no color.Besides,there were 4 out of 7 inbred lines,which had certain restoring ability,and among them there were one plant in inbred lines R2 and two plants in inbred lines R8 that had 100.0 % restoration ratio.Thirdly,the major agronomical characteristics of the 3 restoring crossings were better than their parents,and their heterosis was very obvious.

80 A New Cabbage F1 Hybrid —‘Chenggan No.1’

‘Chenggan No.1’is a new cabbage F1 hybrid developed by crossing self-incompatible line Cai(05-01)with self-incompatible line Jinbei(05-02).Its head is round and compact with dark green leaves.Its head height is about 12 cm and the center column length is about 5.0 cm.It is of superior quality with excellent taste.It takes 55-65 days from transplanting to harvest.Its average head weight is around 0.75 kg and mean yield is about 52.5 t·hm-2.It is suitable for autumn cultivation.

82 A New Color Chinese Cabbage Variety —‘Hongshengbai No.2’

‘Hongshengbai No.2’is a new color Chinese cabbage F1 hybrid developed by self-incompatible line Korea 96-2377 as female parent from a variety‘chunbaohuang’in Korea with backcross inbred lines 97-1346 as male parent from a inbred line Hx90-06.It is middle-early maturity.Its growth vigor is strong,the growing period from planting to harvest is about 65 days,It is 41cm in height,outer leaves are light green,core leaves are pale orange,outer leaves are 22,ball leaves are 41.The leaf ball is inverted cone,like overlapping,26 cm in height,22 cm in diameter.The spherical index is 1.14.It is rich in carotene and selenium,crispy and delicious,good commodity.It has high resistance to virus,resistance to downy mildew.It is suitable for autumn sowing or cultivation in protected field in northern China.

85 A New F1 Hybrid of Pumpkin —‘Ganhongli’

‘Ganhongli’is a high quality F1 hybrid of pumpkin(Cucurbita maxima Duch.ex Lam.)with early maturity and high yield.It was cross-bred by taking YN0112 as female and Jar0521 as male.The vine is short and the fruit setting vine is 40-50 cm in length.The first female flower usually sets on 7-9 node.The fruit is of round and flat shape with bright orange rind and deep orange flesh.Its average fruit weight is about 1.0 kg.It tastes floury and sweet with chestnut flavor.It takes 90 days from sowing to harvesting. Its yield is 37.5 t·hm-2.It is resistant to powdery mildew and virus disease.It is suitable for cultivation both in open and protected field in spring.

88 A New Gourd F1 Hybrid for Protected Cultivation —‘Zhepu No.6’

‘Zhepu No.6’is a new gourd F1 hybrid for protected cultivation,developed by crossing inbred lines G7-4-3-1-2-1 as female parent with G11-3-5-7-1-1 as male parent.The plant grows vigorously,matures early,and is highly tolerant to low temperature,weak light and salt stress.It has good fruit setting.The tender fruit is yellow-green,with long stick shape and round pedicel.The commercial fruit is about 30-40 cm in length,5 cm in diameter.Its single fruit weight is about 0.4 kg with good quality and high amino acid content.It is resistant to Fusarium wilt,middle resistant to virus disease and powdery mildew.It has wide adaptability and is suitable for cultivation in protected field in early spring,and also fit for open field cultivation in late autumn.
