17 Research Progress on miRNA Related to Tomato Response to Abiotic Stress

MicroRNA(miRNA)are endogenous non-coding single stranded small RNA molecules with
19-25 nt length in eukaryotes.They regulate the gene expression mainly by interfering the accumulation of
protein products of target mRNA or degrading target mRNA,and play an important role in plant response to
stress,hormone regulation,reproductive development,and organ differentiation,etc.Plant growth needs
suitable environment conditions.When the environment fluctuation exceeds the tolerance range of plants,
plant growth and development will be harmed.In recent years,along with the further going deep in research
on miRNA,more and more miRNA involved in tomato response to abiotic stress have been discovered.This
paper reviewed the functions of miRNA exerted in tomato growth and development under stress conditions,
including temperature,water,mineral nutrition,light and oxygen,etc.,so as to provide reference for
further research on the regulatory network of miRNA in tomato.

26 Studies on Tolerance of Different Resistant Tomato yellow leaf curl virus Genes to Tomato chlorosis virus

Tomato yellow leaf curl virus(TYLCV)and Tomato chlorosis virus(ToCV)are the 2
viruses,harming tomato more serious in recent years.This experiment made use of 11 tomato materials
carrying different genes resistant to TYLCV;and carried out studies on ToCV tolerance under co-infection of
TYLCV and ToCV.The results showed that there were differences in tolerance to ToCV by tomato materials
carrying different TYLCV resistance genes.Only materials containing single TY-2/ty-2 resistance gene showed
certain tolerance to ToCV,but their plant height and number of fruit set per plant were greatly affected.
However,tomato materials containing TY-1/ty-1TY-2/ty-2TY-3/ty-3 resistance genes at the same time
had better resistance to ToCV.Their disease indexes were below 43.5,and their plant growth potential,fruit
setting rate and fruit maturity were less affected.The CP gene of ToCV and DNA-A whole gene of TYLCV
were cloned and analyzed,respectively.The results showed that the CP gene nucleotide sequence of ToCV
at the time of compound infection was over 99% homogeneous with domestic isolates,and DNA-A gene
nucleotide sequence of TYLCV was over 98% homogeneous with domestic isolates,indicating that no new
viral strains occurred in TYLCV and ToCV at the time of compound infection.

32 Analysis of Genetic Diversity in White Radish Inbred Lines by SSR Marker

In order to verify the genetic background of white radish parent and improve the utilization
effeciency of radish breeding material,this experiment carried out genetic diversity analysis on 42 inbred lines
of white radish,using 38 pairs SSR core primer.The results indicated that the number of primers contained
in chromosomes 1-9 were 2,3,4,6,9,2,3,6,3,respectively.One-hundred and twenty-three
allelic variations were detected among 38 pair primers.The average allelic variation number per locus was
2.86,variating from 2-5.The variation amplitude of polymorphism information content(PIC)was 0.21-
0.70.Average figure was 0.52.Among 42 white radish inbred lines,allele numbers of RSS2114 reached
maximum-5.PIC value was 0.68,showing better primer polymorphism and variety discrimination ability.
Hierarchical clustering analysis indicated that the variation range of genetic similarity coefficient between 42
white radish inbred lines was 0.58-1.00.When the genetic similarity coefficient was 0.61,these 42 materials
could be devided into 3 groups.Group 2 could be devided into 5 sub-groups,and group 3 into 2 sub-groups.
Agronomic traits analysis was conducted on the hybrid progenies from the same or different groups.It was
discovered that the yield superiority of cross between group was significantly higher than that within group.
Besides,the genetic relationship of inbred line with tolerance to bolting was closer.It is required to introduce
new germplasm resources to expand the genetic background of this type parent.

39 Molecular Identity Card Establishment for Broccoli(Brassica oleracea L. var. italica Plenck)Accessions Using SSR Markers

In order to rapidly identify and utilize broccoli germplasm resources,taking SSR molecular
marker fluorescence detection technique,this experiment conducted allelic diversity and clustering analysis
on 22 broccoli main cultivated varieties;screened out the best prime combination;and established
molecular identity card(ID)for these 22 broccoli varieties.According to the number of alleles amplified
and polymorphism information content(PIC),this experiment gradually increased the number of primers
starting with one pair of primers,and screened the primer combinations being capable to distinguish all testing
varieties.Finally,4 pairs of core primer were determined,including BoGMS0501,BoE718,BoGMS0941
and BoE723.Digital assignment was conducted on 4 SSR markers in allelic variation in a variety to
establish molecular identity card for 22 broccoli varieties.It is a simple and practical way to establish
molecular identity card for broccoli accessions using SSR marker.The accessions information is concise and
straightforward.This method has provided technical support for authenticity identification and cultivar-rightprotection
of broccoli accessions.

46 Identification and Evaluation of Common Bacterial Blight Resistance in Common Bean Core Germplasm of Shanxi Province

In order to screen and utilize the common bean resources resistant to common bacterial
blight,this paper identified and evaluated the disease resistance of 151 accessions core germplasm of common
bean in Shanxi by puncture inoculation method on the opposite true leaves,and conducted cluster analysis on
resistant germplasm.The results showed that puncture inoculation of the opposite true leaves could be used
as a rapid and effective method for identification of disease resistance.Through identification,there were 35
resistant and moderate resistant germplasms out of 151 resources,accounting for 23.2% of the total,and 116
susceptible or highly susceptible germplasms,accounting for 76.8% of the total.The only disease resistant
germplasm ‘F0001696’could be used as a high-quality parent material for the improvement and creation
of red kidney bean.Clustering analysis divided the 35 moderate resistant or above core germplasms into 3
groups.The first group included 19 germplasms with short growth period,dwarf plant height and mainly erect
determinate growth.The second group included 4 germplasms with longer growth period,tall plant height
and indeterminate trailing growth.The third group included 12 germplasms with long growth period,medium
plant height and trailing or semi-trailing indeterminate growth.

53 Screening of Sweet Pepper Germplasm Resources Resistant to Meloidogyne enterolobii

Meloidogyne enterolobii is causing increasingly severe damage to sweet pepper.The gene
widely used in nematode resistance breeding is gradually losing its inhibiting effect to Meloidogyne enterolobii
It is urgently needed to excavate new germplasm resources resistant to Meloidogyne enterolobii.This
experiment inoculated Meloidogyne enterolobii to 31 sweet pepper(Capsicum annuum)grmplasms and
conducted identification on their disease resistance after 60 d.Using the 5 disease indexes of Gall Index,Egg
Mass Index,Disease Index,Reproduction Coefficient and Reproduction Index,and subordinative function
value method,this paper carried out comprehensive evaluation on the resistance of Meloidogyne enterolobii
of these tested germplasm.The screened out sweet pepper germplasm resources with strong resistance were

59 Effects of 24-Epibrassinolide on Seed Germination under Low Temperature and Cold Stress Tolerance of Cucumber Seedling

Taking ‘Zhongnong 106’ as test material,this paper studied the effects of 24-epibrassinolide
(EBR)seed soaking on seeds germination under low temperature and substrate adding EBR on seedling
resistance to low temperature.The results showed that soaking seeds with 0.10 mg · L-1 EBR could significantly
improve the germination potential,germination rate,germination index,vigor index,number of lateral roots
and fresh weight of whole plant of cucumber seeds under low temperature.Addition of 0.10 mg · L-1 EBR in
substrate could significantly improve the quality of cucumber seedlings and strong seedling index under low
temperature stress;and increase chlorophyll content,photosynthetic rate,soluble protein content,proline
content in seedling leaves,autioxidant enzyme activities,and root system activity;and decrease the contents
of active oxygen and MDA,thereby to enhance seedling resistance to low temperature.In conclusion,0.10 mg ·
L-1 EBR could promote seed germination under low temperature and enhance cold stress tolerance of cucumber
seedlings.

67 Effects of Regulating Nutrient Solution Temperature on Growth and Yield of Pakchoi Hydroponically Cultured in Protected Cultivation over Summer

The nutrient solution temperature is of great importance for crop growth in the hydroponic
culture system.The excess temperature of nutrient solution often occurs during the production of over-summer
vegetables.This is an issue that will influence production.Taking 3 common pakchoi varieties‘Xialyu
No.2’‘Jinpin 1 xia’and‘Jingguan No.1 F1’as test materials,this experiment conducted cooling process on
nutrient solution by 2 different cooling materials:ice and ice gel,and studied the effects of different cooling
methods on growth and yield of facility and hydroponic cultured common pakchoi over summer by 3 ways
(T1,heat preservation quilt + 3 L ice gel;T2,heat preservation quilt + 5 L ice gel;T3,heat preservation +
5 L ice cubes).The results showed that when 3 L ice gel or 5 L ice cubes were added into the nutrient solution
barrel wrapped with heat preservation quilt at the initial stage of rapid temperature rising of nutrient solution,
the temperature of nutrient solution could be reduced by 3-5 ℃ and suitable rhizosphere temperature could
be maintained about 5 h,which obviously could promote the growth,dry matter accumulation and yield
of common pakchoi.While,the treatment of heat preservation quilt + 5 L ice gel would drop temperature
of nutrient solution too much,resulting in too big temperature difference between above ground and
underground,being unfavorable for common pakchoi growth.To sum up,2 droping-temperature techniques
including heat preservation quilt + 3 L ice gel and heat preservation quilt + 5 L ice cubes,were very effective in
regulating temperature of nutrient solution in hydroponic culture over summer.

73 Analysis on Root Exudates of Different Cadmium-accumulating Cultivars of Water Spinach

In order to study the differences in root exudates components between 2 typical cultivars of
water spinach(Ipomoea aquatica Forsk.)with cadmium-accumulation,this paper carried out detection and
analysis on root exudates of a water spinach cultivar with low cadmium-accumulation‘QLQ’and a high
cadmium-accumulating cultivar‘T308’ by GC-MS method.The results showed that 22 compounds were
identified in the root exudates of‘ QLQ’ and‘ T308’,respectively,including acids,esters,hydrocarbon,
ketone,alcohols,phenols,hydrazines,etc.There were 13 same chemical compounds in root exudates of
‘QLQ’ and ‘T308’,9 specific compounds in root exudates of ‘QLQ’,and 9 specific compounds in root
exudates of ‘T308’.The relative contents of acids and ketones in root exudates of ‘QLQ’ were higher than
those in ‘T308’,while the relative content of esters,hydrocarbons,alcohols and hydrazine in root exudates
of‘ QLQ’ were lower than those in‘ T308’.There were differences in the species and relative content of root
exudates between 2 water spinach cultivars,which might be related to the differences in cadmium accumulating
capacity between these 2 cultivars.

79 Pathogen Identification of Strawberry Root Rot in Fujian Province

In order to definite the pathogenic bacteria of strawberry root rot(purple red leaf),recently
occured in Fujian Province,and guide the prevention and control of this disease,this study collected 20
samples from various regions of Fujian Province,carried out isolation and purification,and obtained 5
bacterial strains.Based on the morphological characteristics of these strains and their conidia,the 5 bacterial
strains were identified as Pestalotiopsis sp..The rDNA-ITS sequences of these strains were amplified by
selected fungus common primer ITS1/ITS4.The ITS sequences of these 5 strains were the same.Through
homology comparison with NCBI database and phylogenetic tree analysis,we found this sequence had the
highest homology with Pestalotiopsis clavispora(Accession:FJ517545.1).The isolated strains were
inoculated back to healthy strawberry plants.They showed the same disease symptom as in the fields.Same
pathogen could be obtained by re-isolation.The pathogenic bacteria of strawberry root rot in Fujian Province
was identified as Pestalotiopsis clavispora by analyzing morphological character of bacteria strains,genome
ITS sequence analysis and inoculation test.

85 Control Effect of Compound Streptomyces rochei with Fosthiazate on Meloidogyne incognita

In order to provide better control measures for Meloidogyne incognita,reduce the dosage
of chemical pesticides,enhance the stability of biological control,and delay the generation of Meloidogyne
incognita
resistance to thiazole phosphine,this study took M. incognita second-stage juveniles(J2s)as target
and determined the nematicidal activity of Streptomyces rochei ZZ-9 fermentation filtrate mixed with Fosthiazate
by different proportions.The results showed that both Streptomyces rochei bacteria liquid and fermentation
filtrate had certain nematicidal activity.Streptomyces rochei ZZ-9 fermentation filtrate could significantly
strengthen the nematicidal effect of Fosthiazate.When the fermentation filtrate was mixed with Fosthiazate
solution(1.0 μg · mL-1)in constant volume complex,48 hours after of treatment,J2s corrected mortality
was 72.64%,the nematicidal effect of Fosthiazate solution could be achieved.

90 A New Pepper F1 Hybrid —‘Xingshuzhoula No.2’

‘Xingshuzhoula No.2’is a new wrinkling pepper F1 hybrid of medium early maturity,
developed by crossing inbred line‘SH1023-2’as female parent and inbred line‘W1F2-1-1-3-1-1’as male
parent.This variety has compact plant type and strong growth vigor.Its first flower sets on about the 10th
node.The fruit is of long cow-horn shape.The ripe fruit is green in color and the old ripe fruit is bright red
in color.The fruit is about 26 cm in length,about 3.2 cm in width,and about 0.25 cm in thickness.The
single fruit weight is about 46 g.The contents of dry matter,VC,capsaicin were 16.4%,1 616 mg · kg-1
25 260 SHU,respectively.In fields,its resistance to anthracnose,virus diseases,epidemic disease and scab
diseases is similar to that of the contrast‘Xiangyan No.15’.Its yield in early stage is about 17.25 t · hm-2
and the total yield is about 34.05 t · hm-2.It is suitable for cultivation in protected and open fields in Hunan,
Hubei,Anhui,Jiangxi,Henan,Jiangsu and Hainan Provinces.

93 A New Eggplant F1 Hybrid —‘Jinong No.3’

‘Jinong No.3’is a new medium-late maturity eggplant F1 hybrid developed by crossing inbred
line‘96-243’as female parent and inbred line‘96-163’as male parent.It is of compact plant type and has
strong growth vigor.Its first flower node sets on the 10-11th node.The fruit is of long stick shape with shining
dark purple peel and light green pulp.The fruit is about 30 cm in length and about 8 cm in width.The single
fruit weight is about 500 g.The contents of VC is 88.0 mg · kg-1 and total soluble sugar is 2.67%.Its resistance
to Verticillium wilt and blight plague in fields is stronger than that of the contrast‘Guozhuangchangqie-2’.Its
yield in early stage is about 60 t · hm-2,and the total yield is about 165 t · hm-2.It is suitable for cultivation in
open fields over summer in northern China.

96 A New Pumpkin F1 Hybrid —‘Baimi No.9’

‘Baimi No.9’is a new small-fruit pumpkin F1 hybrid of early maturity,developed by crossing
inbred line‘TYMB4C1’as female parent and inbred line‘NBF2’as male parent.Its whole growth period is
about 100 days.The plant has strong growth vigor.Its fruits are mainly set on the main lord tendril.The first
female flower sets on the 12-14th node.Its successive fruit setting ability is strong.The fruit is of dumbbell
shape with shallow ridges on peel.The melon is about 30 cm in length,10.8-12.4 cm in width.Tender melon
is dark green in color,and ripen one is orange brown in color.Melon flesh is orange in coor,and about 2 cm
in thickness.Its texture is fine and silky.The soluble solid content is 17.70%.The reductive sugar content is 3.6
g · kg-1.Its quality is excellent.Both matured and tender melons can be harvested.The single melon weight
is about 2.0 kg.Its resistance to powdery mildew,downy mildew and virus disease in fields is equivalent to
that of the contrast‘Gailiangmiben’.It can yield about 60 t · hm-2.This variety is suitable for planting in
Henan Province and Huang-Huai River basin.

99 A New Radish F1 Hybrid of Pigment Extraction Special Type —‘Yunhongluobo No.2’

‘Yunhongluobo No.2’is a new radish F1 hybrid of pigment extraction special type,developed
by crossing male sterile line‘RR05S’as female parent and inbred line‘RR18026’as male parent.It is of
medium-late maturity and takes 70-120 days from sowing to maturity.Its leaves are of pinnate with deep crack
and green in color.The leafstalk is deep red in color.Its fleshy root is of bell shape,and shoulder is circular.
The epidermis and flesh are purple red in color.The fleshy root is about 10 cm in length,7 cm in width.The
single root weight is about 185 g.The radish red content is about 1.6% with even distribution.It is suitable
for procession and extracting pigment.The yield of fleshy root is about 22.5 t · hm-2,and that of pigment is
about 0.375 t · hm-2.In fields,its resistance to downy mildew,soft rot,black rot is equivalent to that of the
contrast‘Tonghaihongluobo’.It is suitable for oversummer cultivation at high altitude(2 000-2 800 m)or
sowing in autumn and winter in southwestern China.
