1 Research Progress on Resistance Breeding of Tomato Late Blight and Molecular Biology

7 Optimization of Technology for Embryo Rescue in Distant Hybridization between Cabbage and Radish

The distant hybridization between dominant male sterile line of cabbage‘C08-11’and radish inbred line‘Q07-12’was made in this experiment.Through in vitro culture of cross-bred ovary and ovule at different developmental stages,the appropriate sampling time,culture medium and additives for the embryo rescue in distant hybridization between cabbage and radish were studied.The results were as follows:ovule culture was obviously better than ovary culture.By ovary culture only 3 hybrid seedlings were gotten and the suitable time for detaching ovary was 9 days after pollination.Adding growth regulators(6-BA,NAA),casein acids hydrolysate and activated charcoal in basic medium B5 was necessary for ovary culture.By ovule culture,29 hybrid seedlings were gotten and the appropriate time for detaching ovule was respectively 14 days after bud pollination and 16 days after florescence pollination.Whether adding growth regulator or not depended on the sampling period and culture medium for ovule culture.

13 Effects of Salt Stress on Chlorophyll Content and Chloroplast Ultra-structure of Different Salt-tolerant Cucumber Varieties

Taking concentration of 225 mmol·L-1 NaCl solution to treat different salt-tolerant cucumber varieties,the effects of salt stress on chlorophyll content and chloroplast ultra-structure were studied.The results showed that under certain concentration of NaCl,at the initiate stage of salt stress,there were not many changes in salt-tolerant varieties.The chlorophyll contents of salt-tolerant and salt-sensitive varieties decreased significantly as salt stress last for a long time,but the decrease range of salt-tolerant varieties is smaller than that of salt-sensitive varieties.At the same time,salt stress could harm the chloroplast membrane system,accelerate the aging of chloroplast,disintegrating gradually the layer,shaping the contour changes,and changing the linage direction of internal layers,and arising a minor expansion,decreasing the number of tablets layers,simplifying the internal structure until total collapse.But the changing degree of salt-tolerant varieties is less than that of salt-sensitive varieties.

17 Primary Studies on Characteristics of Polyphenol Oxidase in Luffa

The characterics of luffa polyphenol oxidase were studied with spectrophotometry using catechol as substrate.The results showed that the optimum temperature of luffa polyphenol oxidase was 20 ℃,and the optimum pH was 6.8.The enzyme activity was higher when the reacting system was at 15~25 ℃ or the pH value was between 6.4~7.2.The experiment of heat stability of luffa PPO indicated that the activity of PPO remained 4.81 % in 100 ℃ by 3 min,while it reduced to 38.08 % in 60 ℃ by 3 min.The dynamics research proved that the Km=0.067 56 mol·L-1,Vmax=0.238 5 A410·min-1 in Michaelis.Menten equation when catechol was substrate.The single inhibitor VC and sodium thiosulfate had inhibiting effect on polyphenol oxidase,but had different effective inhibiting concentration.The effective concentration of ascorbic acid was 50.0 mg·L-1,which was the lowest in the 3 inhibitors,sodium thiosulfate was 100 mg·L-1.Critic acid was the most ineffective inhibitor.

23 Differences in Cytological Development Period of Capsicum’s Microspore with Same Flower Organ Morphology of Different Genotypes

In this experiment,anthers culture of six genotypes capsicum with different fruit shapes and flavors were stained by PIHCH and observed to study the differences between microspore stages and the same shapes of buds.The results showed that the different genotypes had differences between microspore stages and the same shapes of buds,but different genotypes had common characteristics when petals were 0.2 mm longer than sepals in length,and the color from tip anthers to 1/3 back was slightly purple,and the majority of microspores of various materials were at the stage of single nuclear.

23 Effects of Different Spectra on Growth and Nutritious Quality of Radish Sprouting Seedlings

Effects of different spectra of light emitting diode(LED)on growth of radish sprouting seedlings were studied.Light generated from LED and fluorescent lights were used as the control.From the experimental results,red LED was the most effective for the growth of radish sprouting seedlings,and exhibited the highest hypocotyl length,cotyledon area,fresh weight and dry weight.Red LED,combination of red and blue LED significantly increased the contents of soluble sugar and starch.The results suggested that application of red LED could accelerate the growth of radish sprouting seedlings,increase yield and improve partial nutrient quality.

33 Effects of Using Biogas Residue as Substrate on the Growth and Yield of Tomato in Greenhouse

This experiment using biogas residue and coal cinder to study their effects on the growth and yield of tomato in greenhouse.The results showed that the physical and chemical properties of the mixed substrates could meet the requirement of tomato plant growth.Biogas residues could accelerate tomato growth,improve its yield and quality.Among all treatments,the treatment of 3 biogas residue+1 coal cinder(V/V)had achieved the highest root activity and the highest yield(per plan of the first three fruit cluster yields 0.93 kg)and the highest ratio of sugar to acid(6.24).

37 Comparative Test of Qualities between Traditional Excellent Tomato Varieties and New Tomato Varieties

12 excellent new and old varieties were selected from 3609 tomato resources to be conducted comparative test.The results indicate that new tomato varieties are better than traditional excellent tomato varieties on its high uniformity,disease resistance,and good commodity character,while traditional excellent tomato varieties are better than new tomato varieties from nutrient content point of view.If adopting appropriate cultivation measures,it can improve the quality of tomato from its taste and flavor.

41 Virus Detection of Necrotic Stripe Disease by ELISA and Resistance Identification of Processing Tomato Cultivars in Shihezi Area

315 field samples of processing tomato showing necrotic stripe symptom were tested for virus by ELISA using monoclonal antibodies(MAbs)against Tomato mosaic virus(ToMV),Broad bean wilt virus(BBWV)and Cucumber mosaic virus(CMV),respectively.The result showed that the invading rate of ToMV,BBWV and CMV was 69.3 %,66.7 % and 75.1 %,but sole infection rate of these 3 viruses was only 1.5 %,2.4 % and 8.5 %,respectively.These indicated that the mixed infection was probably the main cause to induce necrotic stripe disease in Shihezi area.Situation of the above mentioned 3 viruses carried by 21 processing tomato cultivars were tested by ELISA.The result showed there existed definite differences in resistance among cultivars,although all tested cultivars had relatively high amount of viruses.

46 Diversity Analysis of Fusarium Species from Greenhouse Soil of Melon Crops with UP-PCR

Universally Primed PCR(UP-PCR)was conducted to analyze 25 strains of test Fusarium isolates and 3 strains of positive control Fusarium isolates.The results indicated that a total of 73 bands was amplified using 6 primers,of which 66 bands(90.4 %)were polymorphic.The isolates were clustered into eight groups at the similarity of 0.736 by cluster analysis,among which,14 isolates of F.oxysporum were clustered into one group.

51 Evaluation on the Effect of 50 % Cyromazine SP on Controlling Kidney Bean Liriomyza sativae

The efficacy of 50 % cyromazine SP to Liriomyza sativae in the kidney bean field were investigated in 2007 and 2008.The results showed that control effects of 50 % cyromazine SP were 85.34 %-88.02 %,91.00 %-92.01 % and 92.59 %-95.06 % 3-10 days after spraying 75.0 g,112.5 g,150.0 g effective insecticide per hectare,respectively.Control effects of 50 % cyromazine WP were 90.01 %-93.71 % 3-10 days after spraying 150.0 g effective insecticide per hectare.There were no significant differences in control effects between high dosage of 50 % cyromazine SP and 50 % cyromazine WP.The results of 2007 and 2008 didn’t find significant differences in control effect,and basically identical with those of four years and eight years ago.It confirmed that 50 % cyromazine SP was excellent for effective control of Liriomyza sativae in the fields.

55 Laboratory Toxicity and Field Efficacy of Azadirachtin to Henosepilachna vigintioctopunctata (Fabricius) in Different Instars

Testing result in laboratory showed that the toxicity of azadirachtin to Henosepilachna vigintioctopunctata was deducted with the increase of instars.The larva in 1-2 instar was the most sensitive,both of LC50 values were less than 0.2 mg·L-1. LC50 of azadirachtin to the 3-4 instar larva was less than 1.0 mg·L-1.The toxicity to the adult was decreased obviously,it’s LC50 was 2.122 7 mg·L-1 in 24 h.In the field,the effect of azadirachtin against larva was over 91.17 % at the 7th day at 400-800 times,it suggested the insecticide could be applied at 600-800 times in farming practice,the effect of azadirachtin against adult was over 81.20 % at the 7th day at 400-600 times,it suggested the insecticide could be applied at 400-600 times.

59 Initial Report about Studies on ULV Spray Application of Lignite Extract for the Control of Aphis gossypii Glover

Experiments were carried out for controlling Aphis gossypii Glover of cucumber with lignite extract in greenhouse.The results show that lignite extract with dosage of 10.5-21.0 L·hm-2 which sprayed with high power electrostatic spray system every 6-8 days can significantly reduce the quantity of Aphis gossypii Glover.Lignite extract with dosage of 21.0 L·hm-2 can make the quantity of Aphis gossypii Glover maintain extremely low lever in cucumber growth period.

63 Application of Calcium Soil Amendment in Greenhouse Tomato Production

Lab incubating and field experiments were conducted in order to study the influence of calcium soil amendment on soil properties of saline soil,yield and quality of tomato produced in greenhouse.The results showed that the dormant calcium soil amendment could change the ratio of exchangeable Ca2+ and Na+,decrease exchangeable Na+,pH,EC and total salt contents.The optimized applying rate was 1.50 mg·kg-1.Results from field experiment showed that increasing application of calcium soil amendment could notably improve the yield and quality of tomato produced in greenhouse.Compared with the control treatment,the yield of tomato was increased by 18.7 % and the soluble solid content increased by 20.2 %,titratable acid reduced by 23.1 % and exchangeable calcium content was increased by 34.0 %.The soil physic-chemical characteristics in greenhouse is evidently improved.

68 Influence of Different Calcium Nutrient Solution on Fruit Development of Processing Tomato and Its Quality

In this study,through application of 3 different calcium nutrition solutions on processing tomato variety‘Nonghong No.2’at fruiting period,the results indicating that the use of calcium nutrition solution can improve its colour,and significantly improve the average fruit weight and its lycopene content of processing tomato.Of these,the Leiliyegai can significantly affect the fruit weight,while the Gaibao can greatly affect the lycopene content.Gaibo has the same effect on‘Nonghong No.2’,‘Jiahe No.9’and‘Lige’er 87-5’.

71 A New Chinese Cabbage F1 Hybrid —‘Jinbaicai No.6’

‘Jinbaicai No.6’is a midlate maturing Chinese cabbage F1 hybrid,developed by crossing an inbred self-incompatible line self-crossed from‘Hetou’ (a kind of farmhouse breed in Liaoning Province)with a line self-crossed from‘Tianjinhetaowen’.Its growing period is about 85 days.The hybrid is characterized by its vigorous growth,compact plant type and small stretch.The outside leaves are dark green while the stem is light green.The plant is 56 cm in height.The head is a cylinder with compact twisted central part,53 cm in height,22 cm in width,52 cm in stretch and about 4 kg in weight.Over 75 % of it is eatable and it tastes good.It can be stored for long time and is resistant to TuMV and soft rot as good as‘Jincai No.3’,has strong resistance to downy mildew than‘Jincai No.3’.The average yield is about 120 t·hm-2.It can be cultivated all over China.

73 A New Mini-watermelon F1 Hybird —‘Xinyou No.45’

‘Xinyou No.45’is a new F1 hybrid mini-watermelon variety bred by inbred lines LL-8 and LL-13.It is an early maturing variety.In open fields,its growing period is about 80 days,and fruit growing period about 28 days,In protected fields,it takes 105 days from seedling to harvest.The plant grows vigorously,and the fruits set evenly.The fruit shape is tall and round,and the shape index is 1.15.The peel is dark green with darker stripes and the flesh is light red.The soluble solid content is about 12.0 % in the middle.The weight of average fruit is 2.0 kg,while the yield is 30 t·hm-2 in open fields ,and 60 t·hm-2 in protected fields.It has stronger resistance to powdery mildew than‘Mengtuoling’.

76 A New Parthenocarpic Eggplant F1 Hybrid —‘Liaoqie No.15’

Parthenocarpic Eggplant‘Liaoqie No.15’is a new F1 hybrid developed by crossing inbred line EY3-2-14-5 and EY2-2-6-7.It has mid-early maturity.The growth periold is 113 days.It is of upright plant type,with purple stem,linear clavate fruit and dark purple fruit peel of high gloss.The average fruit weight is 178 g,VC content is 80.0 mg·kg-1,total soluble sugar content is 27 g·kg-1.It has stronger resistance to Verticillium wilt and Phytophthora boehmeriae than‘Liaoqie No.4’,The yield is 73.5 t·hm-2 and suitable for cultivation in protected fields in Liaoning Province.

‘Luyunuo 2008’is a new waxy maize F1 hybrid using inbred line H00-1 as female parent and YH03-15 as male parent.It is middle maturity.The spike is 16.93 cm in length,4.88 cm in width,kernels per row is 31.6,100 grain mass is 35.83 g,seed-producing percentage is 61.30 %,lodging percentage is 2.20 %.It is good both for fresh-eating and processing.The average content of crude amylum is 67.17 % of fresh exemplar,and all are amylopection.It yields over 15 t·hm-2.It has stronger resistance to primary diseases and pests than‘Yunuo No.7’.It is suitable to be cultivated in Sichuan,Chongqing and areas with similar ecological conditions.
