Studies on Function of Pathogenicity-related Gene CcTLS2 in Corynespora cassiicola of Cucumber |

Abstract In order to explore the effect of CcTLS2 gene in the pathogenesis of Corynespora cassiicola,this paper took the strong pathogenic strain HG14102524 as research object,constructed knockout strain by cloning the CcTLS2 gene of Corynespora cassiicola,and combined with bioinformatics analysis,pathogenicity assay,and differential expression analysis,etc. methods preliminarily determined the effect of CcTLS2 on the pathogenesis of Corynespora cassiicola.Comparing with wild type strain HG14102524,the pathogenicity of gene knockout strain CcTLS2 was lost,colony growth rate slowed down,and sporulation quantity decreased significantly.Relative expression levels of Corynespora cassiicola G protein signaling pathway related genes (A7125,A1402,A0835,A8311 and A2529)in CcTLS2 knockout strains were significantly down regulated.The results showed that CcTLS2 might pass G protein related pathway to affect the sporulation mechanism of Corynespora cassiicola,mycelia growth and pathogenicity,and played an important role in pathogenicity of C. cassiicola.
