Abstract In this paper，in viro induction of autotetropolid tomato（Lycopersicon esculentum Mill.）with colchicine was studied on the basis of building regeneration system，the appropriate induction method of tetraploid tomato was culturing cotyledon and hypocotyl callus with buds in medium with 0.2% colchicines for 48 hour．Some indexes of diploid and tetraploid plant were compared and analyzed．The tetraploid plants were found to have stronger growth vigor．Its total chlorophyll contents in tomato leaves and POD activity and SOD activity and soluble protein content were higher than those of diploid．But its MDA content was lower than that of diploid．
