16 Studies on Screening Potato Germplasm Resources with Resistance to Potato Virus Y by Molecular Marker

In order to rapidly and effectively screen germplasm resources resistant to potato virus Y,and speed up the breeding process of new potato varieties resistant to virus,this experiment screened progenies from 9 potato hybridized combinations of processing type introduced from USA using PVY resistant gene molecular marker;and identified/evaluated the prediction accuracy of molecular marker by PVY artificial inoculation.The results showed that among 852 lines in 9 hybridized combinations,163 lines from 4 combinations contained RYSC3 molecular marker with resistant gene Ryadg,accounting for 19.13%;330 lines in 7 combinations contained YES3-3A molecular marker with resistanct gene Rysto,accounting for 38.73%.Among them,2 kinds molecular marker were simultaneously detected in 42 lines from 2 combinations.The results of PVY resistance identification by artificial inoculation showed that 92.73% of the lines containing molecular marker were resistant to PVY after inoculation.28.57% of the lines without molecular marker showed sensitivity to PVY after inoculation.RYSC3 and YES3-3A molecular markers with PVY resistance genes could more accurately identify potato resources resistant to PVY.These 2 molecular markers were helpful for early selection and breeding of potato germplasm resources resistant to PVY.At the same time,this experiment obtained 10 potato germplasm resources of processing type with high and stable production and resistant to PVY.These results had provided parent materials for breeding new potato varieties of processing type and yet resistant to PVY.

23 Development and Utilization of Functional Marker for Resisting ty-5 Gene of Tomato Yellow Leaf Curl Disease

Tomato yellow leaf curl virus disease is one of the important diseases for tomato.On the basis of analyzing ty-5 gene coding sequence,this experiment designed functional marker of ty-5 gene;conducted genotype identification on tomato resistant inbred line ‘t33-14’ to tomato yellow leaf curl virus(TYLCV),susceptible inbred line t2301 to TYLCV,F2 strain of inbred line ‘t33-14’בt2301’,and 26 tomato materials from different sources;and carried out phenotype correlation analysis.The results indicated that functional marker TySNP could accurately distinguish breeding parents from homozygous genotype of tomato material resistant to TYLCV,homozygous genotype of tomato susceptible materials and susceptible heterozygous genotype in hybrid progeny populations.Different genotypes could correspond better with phenotypes,and be used for effective selection for hybrid progeny.Therefore,functional marker TySNP could be used in tomato MAS breeding,and provided a marker-assisted breeding means for directional improvement of tomato cultivars resistant to TYLCV.

29 Bioinformatic Analysis of Fasciclin-like Arabinogalactan Proteins(FLA)Gene Family in Pepper(Capsicum annuum

Fasciclin-like arabinogalactan proteins(FLAs)is widely present in plants and plays a crucial role in various stages of plant growth and development.In order to fully understand the members of pepper FLA gene family,this paper analyzed the physical and chemical properties,subcellular location,phylogenetic tree,chromosome location,gene structure,conservative motifs and cis-acting elements,etc. of CaFLAs by bioinformatics analysis method.The results showed that 25 members of pepper FLA gene family were identified in pepper genome data,being unevenly distributed on 9 chromosomes and divided into 3
subfamilies.Proteins of most CaFLA gene family members encode were mainly acidic,and located on cell membrane.The members of this family contain multihormonal response,adversity stress response,photoresponse,etc. and cis-acting elements related to plant growth and development.

37 Pathogen Identification and Control of Asparagus Stem Blight in Hangzhou City

During 2018—2020,suspected asparagus stem blight case was discovered in Hangzhou Jiahui asparagus planting base.In order to definit the pathogenic species,this study conducted seperation,purification and identification on collected disease samples.It was definited that the disease was asparagus stem blight.According to pathogenic bacteria morphology character,and comparison results of amplification sequences between universal primer and specific primers,the homology of this pathogenic bacteria and Phomopsis asparagi(Sacc.)Bubak was 100%.It was identified as rhizome mildew of asparagine.The results of biological activity determination indicated that mycelium of this pathogenic bacteria was completely inhibited by 25% pyraclostrobin suspending agent.The inhibiting effect to this pathogenic bacteria was followed by 75% chlorothalonil wettable powder.EC50 was 0.13 μg · mL-1.Pot experiment  verified that 25% pyraclostrobin suspending agent had the best effect on controlling stem blight,reaching 94.56%.It could be used as a highly effective agent for preventing and controlling asparagus stem blight.

44 Effect of Water Stress on Physiological Characteristics of Potato Cultivars with Different Maturities

In order to clarify the impact of soil water deficit on growth and development of potato cultivars with different maturities,this paper conducted pot experiment and studied the effects of 4 soil relative water content regimes(W1,75%-85%;W2,55%-65%;W3,35%-45%;W4,15%-25%)on plant height,relative chlorophyll content,fresh weight of organs,photosynthetic characteristics,yield and water use rate of 7 potato cultivars with different maturities.The results indicated that under all water stress the average falling ranges of plant height and intercellular CO2 concentration of early-maturing varieties were greater than
that of the late-maturing varieties.Under severe drought stress(soil relative water content 15%-25%)the average declining ranges of tuberization number and yield of single plant in early-maturing varieties were all greater than that of the late-maturing varieties;while,the average falling ranges of net photosynthetic rate,transpiration rate and stomatal conductance were all less than that of the late-maturing varieties.Considering various indicators,both early-maturing and late-maturing varieties could be planted under light and moderate drought stress(soil relative water content 35%-85%).Late-maturing potato varieties were more suitable to be cultivated under severe drought stress.

53 Analysis of Potato Common Scab Population Structure and PAI Pathogenic Gene in Heilongjiang Province

This study collected potato tubers infected with potato common scab from 6 cities of Heilongjiang Province(Harbin,Jiamusi,Hegang,Suihua,Mudanjiang and Jixi)from 2018-2020;conducted pathogen isolation on potato samples by dilution method;and tested the pathogenicity of the isolated Streptomyces spp. strains by radish seedling method.This study also carried out classification and identification on pathogenic Streptomyces spp. strains according to morphological characteristics,physiological and biochemical traits,combined with 16S rDNA sequences; and analyzed further the PAI pathogenic gene
composition of pathogenic strains.The results showed that there were 5 pathogenic Streptomyces species in 263 pathogenic Streptomyces strains isolated and identified from 154 potato samples collected in 6 cities,including S. scabies,S. acidicabies,S. bottropensis,S. rochei and S. lavendulae.Among them,S. scabies was the dominant strain,S. bottropensisS. rochei and S. lavendulae were reported as potato common scab pathogen for the first time in Heilongjiang Province.Through conducting PCR amplification on pathogenicity island genes(txtAB,tomA,nec1)of pathogenic Streptomyces spp.,2 types of disease-causing gene,txtAB+/tomA+/nec1+ and txtAB-/tomA+/nec1+ were detected.

60 Effects of New Type Seedling Culture with UV-B Supplementary Light Lamp on Growth,Resistance Physiology and Disease Resistance of Eggplant Seedling

In order to explore the effect of UV-B light supplement lamp in eggplant seedling culture,this paper took eggplant variety‘Hangqie No.1’as test material;set up 3 treatments,respectively including UV-B light supplement(with low-dose UV-B band),ordinary light supplement and no light supplement(the contrast);and explored the effects of different light supplement treatments on eggplant seedling growth and resistance physiology.The paper also studied the effects of different light supplement treatments on eggplant seedling disease resistance by indoor determination of UV-B light supplement on pathogen activity,and conducting experiment on pathogenicity at seedling stage.The results showed that the plant height,stem diameter,leaf area,chlorophyll content and ascorbate peroxidase activity(APX),peroxidase(POD),catalase(CAT),superoxide dismutase(SOD),etc. enzym activities of eggplant seedlings were all significantly different from those of the contrast.Among them,the seedling index under UV-B light supplement was the best.In addition,after UV light supplementation treatment,the disease indexes of eggplant seedlings gray mold,anthracnose,and early blight were reduced sharply.Results also indicated that appropriate UV light supplement inside of greenhouse could effectively inhibit plant excessive growth,increase leaf area,raise sound seedling index,enhance antioxidant enzyme activities of leaf blades resistance,and improve plant disease resistance.

68 Effect of Root Application of Glycine on Growth,Yield and Quality of Cucumber Grown in Solar Greenhouse in Autumn and Winter

Taking dense spines and glossy type cucumber‘21-19’ as test material,this experiment set root application of glycine with different concentrations (0,3‰,5‰ and 7‰)4 treatments to study the effects of glycine on growth,yield,and quality of cucumber in autumn and winter in solar greenhouse.Results showed that root application of glycine treatment could improve net photosynthetic rate,chlorophyll content and antioxidant enzyme activity of leaves,reduce malondialdehyde content,increase fruit yield,and raise soluble sugar and VC contents in fruit,indicating that root application of glycine could increase yield and improve quality of cucumber by improving photosynthetic rate and antioxidant capacity of leaves.In view of economic consideration,root application of 3‰ oncentration glycine had the best effect.

74 Effects of Amino Acid Treatments on Growth,Quality,and Yield of Hydroponic Chinese Chive

The special flavour of hydroponic Chinese chive is inferior to that of soil-grown one.The
reason is the inadequate accumulation of CSO in special flavour precursor substance.In order to promote the special flavour of hydroponic Chinese chive,this experiment added 4 kinds of amino acids participated in CSO biosynthesis,including valine(Val),serine (Ser),cysteine(Cys),and glutamic acid(Glu)to neutrient solution;studied the effects of these 4 amino acids on growth and development,nutritional quality,and special flavour of hydroponic Chinese chive.Experiment results indicated that amino acid treatments had little effect on photosynthetic pigment contents and nutritional quality indexes.Their influences on growth index,yield,and pungency degree of Chinese chive were not exactly the same.Among these amino acids,Glu treatment had the best effect.It could not only significantly enhance the growth of Chinese chive,increase its yield and VC content,but also outstandingly improve the expression of 4 FMO genes in Chinese chive,thus promoting its pungency degree to the level of soil-grown products.In conclusion,adding 1 mmol · L-1 Glu to nutrient solution could effectively improve the quality,increase yield,significantly promote pungency degree,and lifting the special flavour of hydroponic Chinese chive.

81 Far-red Light Effectively Accelerates the Accumulation of Biomass,Leaf Blade Anthocyanin,Total Phenolics,and Flavonoids in Basil Plant

In order to definite the effects of substituting far-red(FR)photons for part of photosynthetically active radiation on plant biomass,leaf blade anthocyanin,total phenolics,and flavonoids,this experiment took greenery basil as research object;kept total flux density as 230 μmol·m-2 · s-1 in a plant factory of artificial light type and under condition of 16h photoperiod;measured the effects of substituting farred(FR)photons for part of photosynthetically active radiation on photosynthesis,morphology,biomass,and contents of leaf blade anthocyanin,total phenolics,and flavonoids contents of greenery basil plant.Results indicated that comparing with the contrast,FR photons treatment increased the above ground fresh weight and dry weight of basil plants by 6%-23% and 4%-28%,respectively,plant height and width by 49%-65% and 10%-17%,respectively.FR photons treatment had significantly increased the contents of anthocyanin in basil whole plant and leaf blades,total phenolics,and flavonoids of basil under partial FR light source.The antioxidant capacity of leaf blades had increased by 21%-44%.While,the net photosynthesis and leaf area of basil plant were not significantly affected.

89 Effect of Chemical Fertilizer Reduction on Yield,Quality and Economic Benefit of Zizania latifolia
In order to investigate the effect of applying chemical fertilizer reduction in Zizania atifolia production,this paper took ‘Wanjiao No.1’as test material,and compared the effects of 4 treatments on agronomic traits,quality,yield and economic benefit of Zizania latifolia,including farmers conventional fertilization 100% chemical fertilizer plus chicken manure(CK),3 chemical fertilizer reduction modes:70.9% chemical fertilizer plus high range bio-organic fertilizer(T1),70.9% chemical fertilizer plus medium quantity bio-organic fertilizer(T2),and 70.9% chemical fertilizer plus medium quantity bio-organic fertilizer and decomposed cake fertilizer(T3).The results showed that comparing with the CK,Zizania latifolia yields treated by chemical fertilizer reduction models(T1,T2 and T3)not changed much.But,the tiller numbers were decreased,and fruit formulation of Zizania latifolia were promoted ahead of time.The single plant weight and Zizania latifolia quality were significantly increased.T3 treatment had the best effect in yield increase and quality improvement.The single plant weight,reducing sugar,VC content of Zizania latifolia and fertilizer efficiency of T3 treatment were increased by 20.7%,62.2 %,47.9% and 8.1%,respectively than those of the CK.

93 A New Tomato F1 Hybrid —‘Jindi 363’

‘Jindi 363’is a new tomato F1 hybrid of mid-early maturity bred by crossing inbred line‘09FP-125’as female parent and inbred line‘FP055’as male parent.It belongs to ndeterminate growth type,with strong growth vigor.The first flower sets on the 8-9th node.It takes about 79 days from field planting to first harvesting.The fruit is of round shape.Its transverse diameter is about 8.2 cm and vertical diameter is about 6.2 cm.The matured fruit is red in color without green shoulder.The single fruit weight is 240-260 g.The contents of soluble solid,lycopene and VC are 3.6%,59.8 mg · kg-1 and 163.0 mg · kg-1,respectively.It is highly resistant to TYLCV,root-knot nematode,TMV;moderate resistant to grey leaf spot,leaf mold.It can yield about 120 t · hm-2.This cultivar is suitable for protected cultivation in early spring at Pingba Region of Sichuan Province.

96 A New Eggplant F1 Hybrid —‘Zixian No.1’

‘Zixian No.1’ is a new eggplant F1 hybrid developed by crossing inbred line ‘16A55’ as female parent and inbred line‘16A60’as male parent.It is of mid-early maturity.The plant has medium growth vigor and is of erect type.The first flower sets on about the 10th node.It takes about 50 days from field planting to first harvesting.Its fruit is of strip shape,about 31 cm in length,and about 5.5 cm in width. The average single fruit weight is 293 g.Its peel is dark purple black in color with good gloss.Its flesh is white-green in color.The VC content is 74.3 mg · kg-1,anthocyanin content is 1.88 mg · kg-1,coarse fibre content is 1.05% in fresh fruit.In fields its resistance to Verticillium wilt is stronger than that of the contrast‘Ziliren’.It can yield about 67.5 t · hm-2.It is suitable for protected cultivation in spring in Liaoning region.

99 A New Dried Pepper F1 Hybrid —‘Tianjiao No.24’

‘Tianjiao No.24’ is a new dried pepper F1 hybrid developed by crossing inbred ine ‘15129’ as female parent and inbred line ‘15139’ as male parent.The plant has stronger growth vigor.The plant is about 96 cm in height.It is of early-medium maturity.From field planting to harvesting is about 90 days.The average fruit number per single plant is 32.4.The old ripe fruit is dark red in color.Its fruit is of linear shape.The fruit is about 22 cm in length,and about 1.5 cm in width.Its piquancy is strong.The total content of capsaicin is 3.29 g · kg-1,and VC content is 176.05 mg · kg-1.It is medium resistant to phytophthora blight,CMV and TMV.The average single fruit weight is 13.1 g.It can yield dried pepper about 5.4 t · hm-2.It is suitable to be cultivated in open fields in Gansu,Xinjiang.

101 A New Watermelon F1 Hybrid —‘Nongtian L5K’

‘Nongtian L5K’is a new medium fruit type watermelon F1 hybrid of developed by crossing inbred line‘LA304’as female parent and inbred line‘LA76’as male parent.It is of medium-late maturity.The whole growth period in early spring cultivation is about 103 days,and the fruit development period is about 35 days.The whole growth period in autumn delayed cultivation is about 81 days,and the fruit development period is about 28 days.The plant has strong growth potential.It is easy to bear melon.Its fruit is nearly of round shape.The melon peel is light green in color covered with dark green sharp tooth-shaped strips.There is small quantity of wax powder.The peel is about 1.0 cm in thickness.The flesh is red in color,no hollow,and crisp.It tastes sweet and refreshing.The soluble solids content in the center is 13.0%-14.0%,and that in the edges is 10.3%-11.2%.The single melon weight is about 5 kg.It is moderately resistant to Fusarium wilt.It can yield about 90 t · hm-2.It is suitable for planting in spring and autumn in protected fields of Henan region.
