14 Research Progress on Genetic Breeding of Cucumber Tolerance for Salt Stress

Salt stress is one of the main abiotic stresses affecting cucumber growth and development.It
is of important significance to carry out studies on cucumber tolerance for salt stress and cultivate salt tolerant
varieties for improving cucumber salt tolerance and increasing use ratio of saline-alkali land.This paper
reviewed the following 4 aspects:evaluation of salt tolerance in cucumber germplasm resources;effects of
salt stress on cucumber growth,development and physiology;genetic analysis and genes mining related to
cucumber salt tolerance and major measures for improving cucumber tolerance to salt stress,etc.Emphasis
was laid on research progress made in cucumber breeding for tolerance to salt stress in recent years.At the
same time,the paper put forward the problems existing in current research on cucumber breeding for tolerance
to salt stress,and prospected the research directions in future.

24 Comparative Transcriptome Analysis of CMV Resistance of Pepper at Seedling Stage

In order to explore the molecular response mechanism of pepper under infection of cucumber
mosaic virus and resistant genes related to this virus,the paper took susceptible material‘Qiemen’and
resistant material‘JJ101’as objectives,and conducted high-throughput transcriptome sequencing
on‘Qiemen’and‘JJ101’ before and after innoculation of CMV,and carried out studies on gene expression
and function by bioinformatics method.The results showed that by using RPKM to calculate gene expression
level,1 158 differentially expressed genes(DEGs)were screened out after ‘Qiemen’and‘JJ101’ were
inoculated with CMV.Among them,643 DEGs were enriched in Gene Ontology(GO).Their functions
were mainly referred to enzyme activity,metabolic process and defense reaction,etc.While 287 DEGs could
be included into KEGG pathways,including plant pathogen interaction,plant hormone signal transduction,
etc. process.Studies discovered that pepper immunity to CMV was an extreme complicated and multi-pathway
coordinated process,jointly participated by protein metabolism,defense response,hormone regulation and the
other pathways,laying a theoretical foundation for further research on pepper resistant molecular mechanism.

32 Analysis of Differentially Expressed Genes of Sugar Metabolism in aga2 Mutant Watermelon Fruit under Low Temperature and Weak Light

In order to explore the regulatory mechanism of low temperature and weak light environment
on sugar accumulation of watermelon,this experiment analyzed the transcriptome data of mutant of alkaline
alpha-galactosidase gene aga2,and found that there were 686 up-regulated genes and 990 down-regulated
genes in aga2 mutant fruits under low temperature and weak light.By GO and KEGG analysis,it was further
found that the up-regulated expression of transcription factors:MYB,NAC and WRKY etc. could resist low
temperature and weak light.While,the down-regulated expression of genes related to sugar metabolism
like galactinol synthase(Cla009222),alpha-galactosidase(Cla022883、Cla019238)etc. induced by
low temperature and weak light may be the main reason for the decrease of sugar content in mutant fruit.
By analyzing the transcriptome data,this experiment excavated the genes related to sugar accumulation in
watermelon responding to low temperature and weak light,and laid the foundation for regulation network of
sugar metabolism in watermelon responding to low temperature and weak light.

40 Effects of Different Topdressing Nitrogen Fertilizer on Quality,Growth,and Nitrogen Use Efficiency of Cherry Tomato in Solar Greenhouse

Aiming at the issue of lacking rational nitrogen fertilizer parameter in producing cherry tomato
under solar greenhouse,this paper carried out studies on the effects of different nitrogen fertilizer topdressing
on the quality and nitrogen use efficiency in Beijing under moderate fertility soil conditions.In this study,
cherry tomato‘Qianxi’and‘Hongyu’were selected as experimental materials.The normal amount of
nitrogen fertilizer was determined according to target yield.Four treatments were set up,described as no
nitrogen topdressing(CK),50% normal nitrogen topdressing(50%N),normal nitrogen opdressing(N),
and 150% normal nitrogen topdressing(150%N),respectively.The results showed that compared with the
CK,nitrogen topdressing could improve the yield and quality of cherry tomato;compared with N treatment,
50%N treatment could increase the contents of soluble solid,soluble sugar,titratable acid,VC and nitrogen
use efficiency in cherry tomato,but there were no significant influence on the plant growth,development and
yield of cherry tomato.Therefore,under the condition of soil nutrients and organic fertilizer basal application
closed to the experimental condition in this study,if taking cherry tomato‘Qianxi’ as cultivated variety and
the target yield for winter-spring of‘Qianxi’ was 45 000 kg · hm-2,the recommended amount of fertilizer were
N 120 kg · hm-2,P2O5 75 kg · hm-2,and K2O 360 kg · hm-2.If taking cherry tomato‘Hongyu’ as cultivated
variety,the target yield for autumn-winter of‘Hongyu’ was 60 000 kg · hm-2,the recommended amount of
fertilizer were N 135 kg · hm-2,P2O5 90 kg · hm-2,and K2O 405 kg · hm-2

48 Effects of GABA and CaCl2 Compound on Growth,Chlorophyll Fluorescence Parameters and Reactive Oxygen of Tomato Seedlings under Salt and Alkali Stress

resistance.While,the synthesis of endogenous GABA is related to the concentration of intracellular Ca2+
Taking the salt sensitive tomato variety‘Zhongza No.9’as test material,and adopting salt alkali stress
concentration(60 mmol · L-1 NaCl + 60 mmol · L-1 NaHCO3) with remarkable different phenotype,this
experiment studied the effect of adding GABA and CaCl2 separately and adding their compound on growth,
chlorophyll fluorescence and reactive oxygen,etc. indexes of tomato seedlings under salt and alkali stress.
The results showed that the growth rates of plant height,stem diameter,root length and number of roots of
tomato,its fresh weight and dry weight were all obviously increased under salt and alkali stress,no matter
the exogenous GABA and CaCl2 were added solely or in combination.The leaf color index was significantly
increased and chlorophyll fluorescence parameter was also obviously improved.At the same time,
the exogenous GABA and CaCl2 separate treatment and compound treatment reduced the reactive oxygen
accumulation of tomato leaves.Compared with the separate treatment,SA + CA6 + GABA5 treatment had a
better alleviating effect on the injury by salt and alkali stress,indicating that the addition of exogenous CaCl2
could promote the ability of GABA in enhancing plants salt and alkali tolerance.

56 Studies and Application of New Simplified Water-saving Cultivation Technology for Chinese Cabbage

In order to improve the simplified water-saving cultivation technology for Chinese cabbage
Brassica campestris L. ssp. pekinensis(Lour)Olsson〕,this paper confirmed the application details of
correlation technique implement.This study observed the effect of adding pesticide on seed germination and
insect pests occurrence at the seedling stage,with different concentrations of phoxim and imidacloprid were
added during the seed encapsulation process separately or simultaneously;investigated the effect of different
seeding rate,different planting patterns on the emergence rate at seedling stage;and surveyed the planting
benefits of different types of Chinese cabbage using precision seeding and simplified planting technology.The
results showed that phoxim(effective ingredient content ≤ 3 g · kg-1)or imidacloprid(effective ingredient
content ≤ 70 g · kg-1)added in the seed encapsulation could reduce the insect pests occurrence at the seedling
stage without affecting seeds germination.The emergence rate of 2 seeds per hole could reach over 88%,and
basically fulfilled the production needs.The emergence rate of drip irrigation was 95%-99%,significantly
higher than that of the traditional furrow irrigation.Compared with the traditional cultivation pattern,
water consumption and labor use were reduced by 10.4%-29.5% and 42.2%-79.0%,respectively by this new
cultivation technology during Chinese cabbage growth period.

62 Glycoalkaloid Content in Tubers of Main Potato Cultivars in China and Effect of Mulching Methods and Processing on Its Glycoalkaloid Content

High performance liquid chromatography-evaporative light scattering detection (HPLCELSD)
method was used to measure the content of tuber glycoside alkaloids in main potato cultivars in China.
This paper also studied the effects of different mulching methods and tuber processing on the content of potato
glycoside alkaloids.The results showed that the glycoside alkaloid content of 51 main potato varieties in China
was 106.28-3 770.50 mg · kg-1(DW).Among them,there were 7 potato varieties whose glycoside alkaloid
contents exceeded the safe value 1 000 mg · kg-1(DW).Comparing with those unprocessed,the glycoside
alkaloid content of potato tubers decreased significantly after baking,which was beneficial for improving the
safety of potato tubers.The content of glycoside alkaloids in potato tubers covered with black plastic film was
the lowest,which was significantly lower than that in open field cultivation.Soil mulching after plastic film
mulching had little effect on tuber glycoside alkaloid content.So black plastic film mulching should be used to
grow potato tubers,thus to obtain tubers with low glycoside alkaloid content.

69 Studies On Correlation Between Peel Hardness and Grain Plumpness of Seed-used Zucchini

This experiment measured peel hardness of 2 seed-used Cucurbita pepo sister lines by shore
hardness tester,and carried out slice observation and grain plumpness measurement.The correlation between
peel hardness and grain plumpness was also analyzed.The results showed that there were extremely significant
differences in pericarp hardness of Cucurbita pepo sister lines.The peel hardness of Q212-6-3-1 with hard
peel was 30.3 HD.The width of epidermis 10 columnar cells was 3 526.7 μm.The epicarp thickness was
3 792.6 μm.The thickness of mid mesocarp was 27 700.0 μm.Its difference with soft peel Q212-6-3-2 was
significant or extremely significant.The hundred-grain weight of hard peel fruit was extremely significant
higher than that of the soft peel fruit.Pericarp hardness and hundred-grain weight presented extremely
significant positive correlation.The harder of the peel,the bigger of the hundred-grain weight,the more
plump of the grain.Therefore,hard pericarp could be taken as a major target trait for seed-used Cucurbita
pepo
breeding.

74 Effects of Different Preceding Crops on Clubroot and Yield of Chinese Cabbage in Autumn

In order to screen out the suitable preceding crops for Chinese cabbage production in autumn in
open fields of northern China,this paper took the long-term planting model at the locality of garlic + Chinese
cabbage as the contrast;studied the effects of 3 different preceding crops including wheat,potato onion,and
soybean on clubroot and yield of Chinese cabbage in autumn.The results showed that taking soybean,potato
onion and wheat as preceding crops could reduce the content of resting spores of Plasmodiophora brassicae in
spring-crop soil.The disease index of clubroot of autumn Chinese cabbage treated with potato onion + Chinese
cabbage were all the time significantly lower than that of the contrast.The overground part fresh,dry weight
and yield of Chinese cabbage in autumn treated with potato onion + Chinese cabbage and wheat + Chinese
cabbage were all significantly higher than the overground part fresh,dry weight and yield of Chinese cabbage
gained 30 days and 40 days after sowing.In summary,taking potato onion as the preceding crop can inhibit
the occurance of clubroot in Chinese cabbage in autumn and increase its yield as well.This technology is
suitable for application in local production.

79 Isolation and Identification of Fruit Rot Pathogen in Pumpkin

Post-harvest fruit rot has brought huge economic loss to pumpkin production.In order to
exploit the pathogen of pumpkin fruit rot in Jiaozhou,Qingdao of Shandong Province,this study isolated and
identified the pathogen by molecular biology and morphology method and according to Koch’s Rule.Results
indicated that 2 kinds of bacterial strain were isolated from the infected pumpkin fruit.They were the pathogens
causing pumpkin fruit rot.They were identified as Fusarium solani and Rhizopus stolonifer,respectively.
This is the first report in China that Fusarium solani and Rhizopus stolonifer could infect pumpkin fruit.

83 Control Effect of Five Biological Pesticides Against Asparagus Stem Blight

The bacteriostatic activity of 5 biological pesticides including Bacillus subtilis,lentinan,
kasugamycin,polyoxin and amino-oligosaccharin against asparagus stem blight pathogen,plant safety
and field control effect on these diseases were determined by growth rate method and stem leaf spray
method.The results of growth rate test showed that lentinan and amino-oligosaccharin had no direct antibacterial
activity against Phomopsis asparagi.The safety test showed that 1 200-2 400 t · hm-2 the Bacillus subtilis WP
could inhibit the growth of asparagus.The results of two-year efficacy test showed that 7 days after the last
application,the differences between the average control effect of high-dose of 5 biological pesticides and the
contrast medicament difenoconazole were not significant.And they all showed better control effect on asparagus
stem blight.Fourteen days after treatment,the field control effects of Bacillus subtilis WP,2% lentinan AS and 5%
amino-oligosaccharides AS were all significantly higher than those of the contrast.The 5 biological pesticides
could all be used for green prevention and control of asparagus stem blight,among them lentinan and aminooligosaccharides
belong to plant inducers,had good control effect and longer effective duration.

88 Studies on Factors Affecting Quality and Safety of Flowering Chinese Cabbage Based on Delphi-AHP

Using Delphi-AHP method,this paper investigated and analyzed various factors affecting the
quality and safety of flowering Chinese cabbage〔Brassica campestris L. ssp. chinensis(L.)var. utilis Tsen
et Lee〕in the whole supply chain from farmland to dining table;and evaluated the importance of each factor
in affecting the quality and safety of flowering Chinese cabbage.The results showed that the quality safety
level during planting period among root base class had the maximum influence on quality of flowering Chinese
cabbage.The top 5 factors affecting quality of flowering Chinese cabbage were production place environment
and facilities qualified rate,disease and pest control level,pesticide and growth regulator residue percent
of pass,preservatives residual percent of pass,storage environment and management standardization.The
weighted were 0.130,0.130,0.130,0.128 and 0.082,respectively.

93 A New Cucumber F1 Hybrid —‘Longlyu No.3’

‘Longlyu No.3’is a new cucumber F1 hybrid developed by crossing inbred line‘JZ161’as
female parent and inbred line‘4F’as male parent.Its plant has medium growth vigor.The fruits mainly
grow on the main vine.The first female flower sets on the 3-5th nodes.The fruit is of straight and rodlike
shape,and is about 30 cm in length with short stalk.Its peel is bright dark green in color with dense spines.
The flesh is green in color.Its VC content is 130.0 mg · kg-1,and soluble solids content is 3.31%.The plant
is resistant to downy mildew,powdery mildew,and blight.It can yield about 57 t · hm-2.It is suitable to be
planted in the Three Northeast Provinces and Inner Mogolia in protected fields in early spring.

95 A New Chieh-qua F1 Hybrid —‘Jibao No.1’

‘Jibao No.1’is a new chieh-qua F1 hybrid developed by crossing inbred line‘Heimao’as
female parent with inbred line‘Lyufeng’as male parent.It is of early-medium maturity.It takes 69 days in
spring and 54 days in autumn from sowing to harvesting,and the harvest period can last for 36 days in spring
and 33 days in autumn.Its fruit is of cylindrical shape.The peel is dark green in color with even distributed
medium-sized white spots,but without furrows.The flesh is compact with superior quality.Its fruit is about
19 cm in length,7.5 cm in transverse diameter,and 1.7 cm in flesh thickness.The single fruit weight is 600-
720 g.The commodity fruit rate is about 95%.Its VC content is 219.0 mg · kg-1,soluble solid matter content
is 36.0 mg · g-1,and crude protein content is 5.16 mg · g-1.It can yield 60.0-67.5 t · hm-2.It is suitable for
autumn cultivation in chieh-qua producing regions of Guangdong Province.

98 A New Chinese Chives Variety —‘Pengbao’

‘Pengbao’is a new Chinese chives variety of developed by crossing a favor
strain‘Guilindaye’as female parent with an inbred line‘Jinyin No.1’as male parent through sexual
hybridization and systematic breeding.It is of erect plant type and about 49 cm in plant height.Its leaf
blades are green in color with 40 cm in length,and 0.7-1.1 cm in width.The single plant has 4-6 leaves.
The average single plant weight is 9.6 g.It has strong growth vigor.It is strongly resistant to cold and will
not go dormant in winter.In fields,its resistance to grey mold is equivalent to that of the contrast‘791’.
Its VC content is 302 mg · kg-1,and β-carotene content is 14.70 μg · g-1.It can be harvested 6-7 times a year
and yield about 195 t · hm-2.It is suitable to be cultivated in open and protected fields in Henan Province and
surrounding areas with similar ecological conditions.

101 A New Fresh Edible Sweet and Waxy Corn F1 Hybrid —‘Baitiannuo 209’

‘Baitiannuo 209’is a new fresh edible sweet and waxy corn F1 hybrid bred by crossing inbred
line‘LN38’as female parent and inbred line‘LN205’as male parent.It is of medium-early maturity.
It takes about 83 days from sowing to fresh ear harvesting.It is of semi-compact plant type.It is 217 cm in
plant height and 105 cm in ear position height.Its ear is of cylindrical shape,19.1 cm in length and 4.5 cm
in ear diameter.There are 14 rows in each ear and 33 grains in one line.Its grain is white in color.The
proportion of sweet and waxy grain is 1∶3.The amylopectin content takes 97.5% of the total starch content.
The hide trimmings rate is 8.1%.It can yield fresh ear about 13.5 t · hm-2.It is resistant to large spot disease
and medium resistant to small spot disease and sheath blight.It is suitable to be planted in Jiangsu Province and
surrounding areas with same ecological conditions.
