The tomato seeds from Turkey were detected by DAS-ELISA and RT-PCR. The results showed that Tomato spotted wilt virus(TSWV)was detected in this batch of tomato seeds. Sequencing results of PCR products showed that the nucleotide sequence similarity of the fragment to the other TSWV isolates was 99%-100%. TSWV was confirmed in this batch of tomato seeds.
The gynoecious line‘ D1104-2-4’,pistillate line‘ D1158♀-2’ and monoecious line‘ D0528-2’ were used as female parents,and the hybrid generation was obtained by crossing with monoecious line‘ D0708- 2’ as male parent.The paper investigated the female related agronomic characters at 3 periods including early blooming stage,fruiting early stage and fruiting period; then screened the significantly different combination to carry out trans-criptional sequence;and finally analyzed the differential genes of MADS-box family among cucumber hybrid combinations.The results showed that the F1 generation with‘ D1104-2-4’ as female parent had super parent advantage on the numbers of female flower and female flower node rate.And F1 of all 3 hybrid combinations had the early maturing mid-parent advantage.The results of transcriptional analysis showed that the F1 gene expression in the whole gynoecious line‘ D1104-2-4’ combination was significantly biased to the maternal parent,and only 6 of 37 cucumber MADS-box family genes were different from the female parent,and 17 of them were differ in expression with male parent.Bioinformatics analyzed and named a significantly differential expre ss gene CsMADS-box AGL62 that encodes 187 amino acids in the whole length of 745 bp.The encoded protein was belonged to an unstable hydrophilic transmembrane protein in the mitochondria.The NCBI alignment results showed that this protein was a transcription factor.Systematic analysis found this protein had the highest homology to AGL62 in muskmelon.In cucumber,this gene was belonged to the same branch with the other cucumber AGL62 subtypes and belonged to type I.
Based on the full length of eight hydrogen phytoene synthase(PSY 1)from NCBI databases, this study designed specific sgRNA according to the PAM(proto adjacent motif)site of the gene sequence, and constructed CRISPR/Cas9 Level 1 and Level 2 vectors. Then the designed vector was transformed to Agrobacterium,by the Level 2 vector were used to transform the tomato cotyledons. The knockout efficiency of sgRNA in tomato genome was detected by PCR product sequencing. The results showed that 22 of the 36 transgenic plants showed a different number of base deletion,increase or interchanges in the targeting area of PSY 1. The gene knockout efficiency was 61% by initial estimates. The technique of CRISPR/Cas9 mediated gene knockout was established in this study. This technique could stably generate gene knockout in tomato.
In order to clarify the feasibility of regeneration and grafting reuse of rootstock and scion after eggplant grafting,this paper took‘ Qiezhen No. 1’ as rootstock and‘ Yuanza 471’ as scion;studied the growth and development process of rootstock and scion residue regeneration seedlings;and compared the healing and growth indexes of residue grafting seedlings with initial grafting seedlings. The results showed that both the residue stock of the rootstock and the scion could be healthily regenerated about 30 days after grafting. The plant stem diameter could reach about 2.5 mm. Compared with the sowing seedlings,the rootstock and scion could shorten the seedling raising time about 45 and 10 days,respectively. Compared with the initial grafting seedlings,the survival rate,transporting capacity,resultant force and net growth of grafted seedlings in 35 days did not have significant differences when grafted with rootstock and scion residue. And the cost of residue grafting seedlings was obviously lower than that of initial grafting seedlings. Counting by every ten thousand grafted seedlings,the production cost could be saved by 696.4 yuan. The grafted rootstock and scion residue could be regenerated and grafted again after grafting. Thus,the economic benefits could be improved.
This paper studied on the effect of applying sulfur-silicon combination on the growth, photosynthetic characteristics and quality of garlic seedling by adopting the method of deep liquid flow hydroponics. The results showed that this combination could improve the plant height,cauloid width and length of garlic seedling,and also enhance the photosynthetic pigments content and photosynthetic parameter. The most prominent effect was found in concentrations of 2 mmol·L-1 sulfur with 1.50 mmol·L-1 silicon. Suitable concentration of sulfur and silicon combination was in favor of increasing the contents of allicin,soluble protein,soluble sugar and VC. The best effect could be gained in the concentration of 2-3 mmol·L-1 sulfur combined with 0.75-1.50 mmol·L-1 silicon. In conclusion,the concentration of 2 mmol·L-1 sulfur combined with 0.75-1.50 mmol·L-1 silicon was the most beneficial for the growth and quality of garlic seedling by hydroponic culture.
This paper analyzed the cadmium contents in different parts of oriental melon and fruit quality through cultivating melon on the land with cadmium polluted soil.Results showed that in all 12 tested different types of melon fruit,the cadmium contents were lower than that set by The National Food Safety Standards,Limit of Contaminants in Food,GB 2762—2012 requested the cadmium content in melon fruit was 0.05 mg·kg-1. The cadmium content in fruits of IVF-28 was the lowest,only 0.015 3 mg·kg-1.The cadmium distribution in melon plant was:base stem and leaf > root > upper stem and leaf > fruit.Integrating cadmium content and fruit quality traits,we screened out IVF-28 suitable for planting in moderate and below heavy metal polluted areas.
This paper conducted investigation at a continuous cropping land for many years in Xiao County of Anhui Province on the main agronomic traits and disease level indexes of Alternaria leaf blight during 3 growth periods(T1,T2 and T3,70,90,110 days after sowing)of 160 carrot accessions from abroad and at home. The results showed that the disease level indexes of Alternaria leaf blight increased gradually with the growth of carrot and there were significant differences between the 3 periods. The average disease level indexes were 0.9, 1.8 and 2.1,respectively. Based on T3 index,160 accessions could be divided into 5 groups,and group I had the strongest resistance including 21 accessions. Their disease level indexes were 0.25-0.75. The upper ground main agronomic traits of group I were also significantly better than those of the other groups.
According to the characteristics of clubroot occurrence in broccoli,this paper studied the control effect of 50% fluazinam suspension(SC)on clubroot and safety for plant growth in seedling and adult-plant stages by 2 different applying methods:one was homogeneously stirred seedling substrate while sowing,the other was root-irrigation at different dates after transplanting in field.The results showed that the growth of seedling was inhibited severely by 3 concentrations of 50% fluazinam SC.Compared with the control group,the plant height became shorter,and first true leaf became smaller.The higher the concentration was,the more obvious the inhibition was.So fluazinam was not suitable to be used in the seedling stage.Meanwhile,the rate of emergence was not significantly affected.The control effect was variant,as applying 50% fluazinam SC irrigating root after transplanting in field at different dates.The relative control effect of treatment with root-irrigation at 3 days and 30 days after transplanting was extremely the best,reaching 92.21%,which was extremely significantly superior to the other treatments.The second way was conducting root- irrigation at 3 days after transplanting.The relative control effect was 74.03%,which was extremely significantly better than that after disease onset.The control effect of the treatment of 2 times was obviously better than that of the treatment of 1 time.Meanwhile,the control effect of treatment before symptom appeared was better than that of the treatment afterwards.
Pesticide preparation form has great effect on its control effect,duration efficacy and residual in environment.In order to understand the optimum application amount and safety interval of rotenone,this study analyzed the degradation dynamics and terminal residue of 2.5% rotenone microcapsule suspension on pakchoi and in soil by high performance liquid chromatography(HPLC).The results showed that the degradation dynamic equations of rotenone on pakchoi and in soil fitted the first-order reaction dynamic equation.Their half-lives were 2.58-2.74 days and 2.88-3.07 days,respectively.When 2.5% rotenone microcapsule suspension was applied at 1.50 kg·hm-2 or at 2.25 kg·hm-2,the final residual concentration of rotenone on pakchoi and in soil was conformed to the residual requirements and it is safe to use.
‘Zhongnong No.33’ is a new cucumber F1 hybrid developed by crossing inbred lines‘ 12173’ and ‘09676’ as maternal parent and paternal parent.It grows vigorously with high tolerance to low temperature and weak light.Its fruit has good commodity and is shining dark green in color.The fruit is about 35 cm in length, about 3.4 cm in diameter and its neck is about 4.5 cm in length.The fruit has small chamber and light green flesh.There are many white spines and small tumours on its peel.It is highly resistant to WMV,medium resistant to CGMMV,Fusarium wilt and scab.It is of early maturity and has sustainable fruiting ability and obvious high yielding potential.Its yield can reach 150 t·hm -2 in solar greenhouse under long season cultivation. It is suitable for cultivation in solar greenhouse and under shelter in spring.
‘Gantiannuo No.2’ is a new fresh sweet waxy corn bred by crossing inbred line‘ Nuo J11’ as female parent and‘ Tiannuo 7’ as male parent.Its plant height is 231.0 cm,and ear height is 110.0 cm.Its ear is of conical shape.It has wide adaptation.Its kernel is stick and sweet.Its peel is thin.Its quality is superior. Its fragrance is pure.It is moderately resistant to common smut,highly resistant to stalk rot and red leaf disease. It can yield about 18 t·hm -2.It takes about 102 days from planting to harvesting.It is suitable to be planted in spring maize area of Northwest China.
Abstract:‘Jinsong 66’ is a new F1 hybrid autumn loose-curd cauliflower of medium maturity developed by crossing male sterile line‘ D-16’ as female parent and inbred line‘ M-13’ as male parent.It takes about 70 days from field planting to harvesting.Its plant has strong growth vigor.Its ball is loose,tender,smooth with green stem.The average single ball weight is about 2 kg.It can yield about 60 t·hm -2.It is suitable for cultivation in Beijing,Tianjin,Shandong,Hebei,Henan in open field in autumn.
‘Sumeng No.6’is a new watermelon variety of small fruit type with outstanding quality.It is developed by crossing‘ G42’ as female patent and‘G38’as male parent.Its whole growth period is 105-117 days and the fruit development period is 30-35 days in early spring protected field cultivation.The fruit is of round shape,and its peel is green in color with dark green stripes.Its peel is 0.44 cm in thickness.The flesh is red in color with crisp texture.Its central sugar content is 12.3%.The single fruit weight is 1.5-2.0 kg.It can yield 30- 48 t·hm -2.It is resistant to Fusarium wilt.It is suitable for spring protected field cultivation in Jiangsu Province and Beijing area.
