23 Effect of Different Storage Temperature on Transplant Quality of Broccoli Plug Seeding
PAN Xiao-Bing, TIAN Yong-Qiang, LI Juan-Qi, YANG Tian, GAO Li-Hong
To study the effect of different storage temperatures on broccoli(Brassica oleracea L.var. italica Plenck)seedling quality, growth and development after fix planting, this paper screened the related morphological indexes of seedling new heart leaf growth rate after planting, taking‘good’as test material to evaluate the quality of broccoli seedlings under temperature 11,16,21 ℃ and stored for 0,2,4,6 days, respectively. The results
showed that in the storage temperature of 11 ℃,seedling’s morphanological index maintained in good shape, the chlorophyll content per unit area was relatively higher, and the seedling new heart leaf grew rapidly. The correlation analysis results showed that the sound seedling index and plant recovery ability after fix planting during storage period had the highest correlation coefficient. The sound seedling index could be an indicator for evaluating
the storage quality of the broccoli seedling. The path analysis results showed that composite indexes of chlorophyll content per unit area, sound seedling index, specific leaf weight could well reflect the growth recovery ability of broccoli seedling after short-term storage.

28 Cloning and Expression of SRK2F Gene in Chinese Cabbage
ZHOU Zhi-Guo, WANG Cong-Yan
Taking non salt and drought tolerant inbred lines of Chinese Cabbage〔Brassica campestris L.spp. pekinensis(Lour)Olsson〕SY-14-06 as material, extracted total RNA from root, and reverse transcribed cDNA.According to SRK2F gene sequence of Brassica rapa, primers were designed and 1 044 bp open reading frame(ORF)was cloned. The SRK2F protein contained 347 aminoacids, with a prediction molecular weight of 39.3 kD and PI of 4.88.We construct the procayotic expressive plasmids pEASY-E1-SRK2F using pEASY-E1 vector. After transformation to Transetta(DE3),the expression of recombinant proteins were detected with SDSPAGE.The structural analysis of SRK2F though Smart-embl showed that it contained silk threonine structural domain, which was located at the position of 4-260 aminoacid residues. The ClustalX2 comparison indicated that the SRK2F had close genetic relationship with Arabidopsis thaliana. Finally, purified this protein by affinity chromatography medium, which was known as Nickel ion metal affinity chromatography medium. Thus, gained the purified fusion protein.

33 Effects of Inoculations of Glomus mosseae and/or Rhizobium on Potted Bean Growth
LI Ya-Hui, GAI Jing-苹, CHEN Qing, LI Zhi-Fang
Under greenhouse condition, common bean(Phaseolus vulgaris Linn.)‘Taiguowujindidou’was sowed in pots to estimate the effects of different inoculation ways on bean’s plant growth, nodulation, total pod weight of single plant,AM colonization rate, nitrogen and phosphorus contents and absorption of bean plant. When seeding, the beans were inoculated with Glomus mosseae(AM),rhizobia(RH),Glomus mosseae and
rhizobia(AM+RH),and the beans without inoculation were taken as the control(CK). The results indicated that dual inoculation(AM+RH) could obviously promote the plant growth of beans. Compared with inoculation with Glomus mosseae(AM),rhizobia(RH)and none(CK),the plant total dry weight of the beans inoculated with AM+RH increased 34.3%, 82.3% and 57.9%, respectively. Besides, the dual inoculation could increase the total pod weight of single plant and promote bean’s absorption of nitrogen and phosphorus. The phosphorus absorption of total plant was increased by 42.1%, 314.8% and 325.4%, respectively than that by AM,RH and CK. While, the nitrogen absorption of total plant was increased by 36.8%,45.1% and 43.4%,respectively than that by AM,RH and CK. The Glomus mosseae and rhizobia had synergistic effect on common bean, and the Glomus mosseae could effectively improve bean’s resistance, reduce the yellow leaf ratio, and thus decrease the occurrence of diseases.

LIU Jie, WU Feng-Zhi, ZHOU Xin-Gang
This paper studied the effects of microorganism population in continuous and rotated cropping on cucumber seedling growth and soil enzyme activity. The results showed that in rotated cropping microbial treatment, the fresh weight, chlorophyll content, the activities of neutral phosphatase and invertase of cucumber seedlings were significantly higher than that in continuous cropping microbial treatment. Both the continuous and rotated cropping microbial treatments had increased the fresh and dry weights, chlorophyll contents, total leaf area of cucumber seedling, and significantly increased the activities of soil neutral phosphatase, invertase, urease. Summing up the above, soil microorganism population in both cropping systems had a positive effect on the growth of cucumber seedling, and the positive effect on soil microorganism population in rotated cropping was stronger than that in continuous cropping.
