LI Fei, ZHANG Shu-Jiang, ZHANG Shi-Fan, ZHANG Hui, SUN Ri-Fei, ZHANG Zhen-Xian
Taking Chinese cabbage〔Brassica campestris L. ssp. pekinensis(Lour)Olsson〕‘Jihong 82’,some of #534DH and #438 population as testing material, studies the effect of mechanical extraction and artificial extrusion on micro-spore viability and extraction quantity. The result indicates that mechanical extraction can replace the artificial one,get multiple samples during short period of time, and rapidly adjust the concentration of microspores culture.40 suitable buds are collected in 5 mL centrilfuge tubes,3 000 r·min-1 micro-spores gained by 10 s smashing extraction are isolated in 25 mL NLN-13 medium.Thus,micro-spore concentration is insured in suitable culture scope, and successfully induced to embryogenesis.

17 Brief Report on Chinese Cabbage Male Sterile Line MS200712
ZHANG De-Shuang, ZHANG Feng-Lan, YU Yang-Jun, ZHAO Xiu-Yun, YU Shuan-Cang, WANG Wei-Hong, SU Tong-Bing, LU Gui-Xiang
Creation and abortive mechanism of new male sterile lines are hot-topics on Chinese cabbage breeding.F1 hybrid crossing Brassica allohexaploid(AABBCC,n=27),which owns A,B and C chromosomes simultaneously, with Chinese cabbage variety of‘Jinchun’was obtained. Then the F1 as a male parent was crossed with Chinese cabbage variety of‘Yihe No. 5’.Thus male sterile MS200712 was founded due to multiparents crossing.MS200712 owned yellow petal and light-yellow anther. The anthers were regressive, which were 100% male sterility.Stigma of MS200712 was normal and nectaries were vigor. Meanwhile 832172,male parent of‘Beijingnew No. 3’was crossed with MS200712,and male sterile MS20071208-916 was obtained.

21 Effects of Arbuscular Mycorrhizal Fungi Inoculation at Different Stage on Yield and Nutrient Quality of Cucumber(Cucumis sativus L.) in Plastic Tunnel
LI Cong, HE Chao-Xing, YAN Yan, LI Yan-Su, YU Xian-Chang, Martina Janouskova Miroslav
This experiment studied the effect of inoculating arbuscular mycorrhizal fungi at different stages on cucumber yield and nutrient quality in plastic film greenhouse. The results showed that the yield of cucumber inoculated with Glomus intraradicese(GI) at seed sowing and transplanting stages could be increased by 54% and 34%,respectively as compared with the non-inoculated control. Inoculation at seed sowing stage, the chlorophyll content in cucumber leaves was significantly increased by 14.7%-17.7%,compared with that of the control. The nitrate contents of cucumber inoculated at seed sowing and seedling transplanting stages were decreased by 9.4% and 15.0%,respectively comparing with the control. While their total sugar contents were increased by 8.9% and 7.1%,respectively comparing with the control. The crude protein content in cucumber inoculated at seedling transplanting stage increased by 4.6% as compared with the control. The VC content in cucumber inoculated both at seed sowing and seedling transplanting stages were increased significantly. These results indicated that GI inoculum might improve cucumber growth, increase its yield and improve its nutrient quality.

25 Characterization of Soils Bacterial Community Structures in Rhizospheres of Tomatoes Infected with Bacterial Wilt and Its Non-infected Plants
YANG Shang-Dong, ZHAO Jiu-Cheng, GUO Yi-Juan, WU Jun, LONG Ming-Hua
A Polymerase Chain Reaction-Denaturing gradient gel electrophoresis(PCR-DGGE)technology was used to analyze the soil bacterial community structure in rhizospheres of tomato infected by bacterial wilt and non-infected plants. The results showed that the indices of diversity(Shannon-Wiener index),richness and evenness of bacterial community in rhizosphere soils of non-infected plants were 2.217,14 and 0.840, respectively. In contrast, the same indices in rhizosphere of tomato infected by bacterial wilt were 1.579,10,0.686(wilting slightly)and 1.571,7,0.807(wilting severely).Meanwhile, the indices of diversity and richness were decreased with the wilting degree of infected plants.

30 Studies on Nutrient Components Change during Chieh-qua Fruit Development
LIU Zheng-Guo, WANG Peng, CHEN Yong
In order to study the dynamic change of nutrient contents in the development of chieh-qua
(Benincasa hispida Cogn.var.chieh-qua How.)fruit, the contents of VC,organic acid, soluble sugar and soluble protein were analyzed taking 5 chieh-qua cultivars‘Nanningmaojiegua’,‘Meihua No.5’,‘ 7-2-1-4-2-1-2-2’,‘GF-7-1-1’ and‘ CK-3-4-3-2-1-3’ as experimental material during 10,20,30,40 days after pollination. The results showed that the changes in nutrient components of VC,organic acid, soluble sugar and soluble protein were basically similar during the fruit development of these 5 chieh-qua cultivars. Among them the VC contents reached the highest level in the early stage, then decreased rapidly, and tended to be stable at the mutured stage. While the organic acid contents showed a trend of decreasing first, then increasing. The soluble sugar content showed a gradually increasing trend. The soluble protein content showed a trend of decreasing first, then increasing slightly. However, there were differences between different cultivars.

33 Studies on Using Maize Straw and Mushroom Residue to Replace Peat as Nursery Media for Tomato and Sweet Pepper Seedlings
DONG Chuan-Qian, YIN Cheng-Cheng, WEI Min, YANG Feng-Juan, SHI Qing-Hua, WANG Xiu-Feng, ZHANG Wei-Li
The maize straw compost, mushroom residue compost, peat and vermiculite were used to composite different complex media for tomato(Solanum lycopersicum L.)and sweet pepper(Capsicum annuum L.var. grossum Bailey)seedling.Comprehensive indexes such as seedling growth, seedling index, physiological activity, indicated that the media of mushroom residue compost+ peat+ vermiculite(1∶1∶1,V/V)had the best effect on tomato seedling, the next was the media of maize straw compost+ peat+vermiculite(1∶1∶1,V/V).The media of maize straw compost+vermiculite(2∶1,V/V)had the best effect on sweet pepper seedling, and the next was mushroom residue compost+peat+vermiculite(1∶1∶1,V/V).The results showed that mushroom residue compost and maize straw compost could improve the physical and chemical characteristics of complex media, promote tomato and sweet pepper seedling growth, and cultivate healthy seedlings with high quality.
