Research Progress in Cucurbit Seed Traits Inheritance and Molecular Biology

Seed traits are closely related to plant growth and development,yield and quality of product organ,and environmental adaptability of plant.It is important to investigate thoroughly the inheritance and development regulation mechanism of seed traits for improving crop germplasm, increasing yield and solving the problem of biological energy s ource.In recent years,great progress has been made in studying Cucurbit seed traits.Studies on inheritance,development regulation mechanism and molecular biology of seed traits were reviewed in this paper. In addition,problems existing in current research were discussed and future research orientation was prospected.

Analysis of Influencing Factors on Melon Production and Operation Benefits —Taking Xinjiang Melon Farmers as Example

Taking 181 households of Xinjiang melon farmers as examples,on the basis of investigation data,this paper carried out descriptive statistical analysis on the factors influencing production and operating statusquo of melon. The paper adopted semilog multivariable linear regression method, and systematically analyzed the factors influencing melon production and operation benefits in Xinjiang,and the correlation between planting scale and benefit.The results indicate that the benefits are influenced by planting pattern,production mode,planting scale and region selection,etc. Accordingly,the paper puts forward suggestions in detail to improve the melon production and operation benefits in Xinjiang.

Construction of Watermelon Genetic Map and Fruit Shape Index QTL Analysis

Taking round shape watermelon line‘ Huayuanmuben’ as female parent and oval shape line‘LSW-177’as male parent,we crossed them and obtained F2 population constructed with 180 single plants.We measured the length and width of the ripened watermelon〔
Citrullus lanatus
(Thunb.)Matsum. et Nakai〕,obtained fruit shape index from the ratio between fruit length and width, constructed the genetic map and analyzed the QTLs of relevant characteristics.The constructed genetic map contains 45 SSR-markers belonging to 13 linkage groups.The covering genome length was 547.3 cM.The average distance between markers was 12.44 cM.The one QTL closely associated with fruit shape index was located in the 3rd linkage group between MCPI-12 and CMN05_69.This QTL was named as Fsi3.1.Its contribution rate was 2.8%,and its additive effect was negative,which showed synergia additive effect in reducing fruit shape index.

Purity Identification and SNP Site Obtain of Chinese Cabbage Hybrids Using Multiplex EST-SSR Marker

In this research,10 Chinese cabbage〔
Brassica campestris
L. ssp.
pekinensis
(Lour) Olsson〕hybrids and their parents were taken as study objects.Based on the EST sequences from NCBI genome data base,a SNP site and multiplex EST- SSR sites,which could be used to identify the purity of Chinese cabbage hybrids,were screened by SSR marker and high resolution melting(HRM) technology.According to the requirement of seed purity identification by high throughput molecular technology,the key techniques such as seedling culture condition of single seed,extraction status of single grain plant, rapid DNA extraction method and establishment of multiplex PCR system were groped and optimized.Results showed that the single germinating seed after 48 h could gain DNA with higher quality by the improved CTAB method in 3 h. Meanwhile,plant leaves reached seeding stage after 7 d,could complete DNA extraction by chexe-100 method within 40 min.The SNP site obtained by screening could be used to indentify the purity of 6 Chinese cabbage hybrids,and the multiplex ESTSSR locus could be used to indentify the purity of 10 Chinese c abbage hybrids.

Cadherin Fragments and Optimization of Its Conditions

cadherin gene fragment.In order to enhance its expression in
Escherichia coli
,we studied the effect of time, temperature and IPTG concentration on fusion protein expression and its solubility.The results showed that the fusion protein gave rise to the maximum expression when host cells were cultured in LB at 37 ℃ for 1.5 h and then induced by 0.1 mmol·L
-1
IPTG at 25℃ 180 r·min
-1
for 36 h.And the temperature change had little impact on the solubility of fusion proteins. SDS-PAGE test also indicated that the molecular weight of fusion proteins was the same as expected,wh ich is about 16 kDa.

Analysis of Genetic Diversity Morphological Marker in Eggplant Germplasm Resources

This study evaluated the level of genetic diversity of eggplant(
Solanum melongena
L.) germplasm resources through making morphological markers on 76 copies of eggplant germplasm resources. The results showed that the morphological traits of the tested eggplant germplasm showed genetic differences. The average variable coefficient between each germplasm traits was 54.83%,of which the largest variable coefficient was leaf spines,493.3%,the least variable coefficient was fruit curvature,10.9%. Clustering analysis was carried out on eggplant phenotypic character,76 eggplant materials were divided into 3 groups taking Pearson correlation coefficient 23 as clustering threshold,if taking Pearson correlation coefficient 19.5 as clustering threshold,76 eggplant materials were divided into 7 sub-groups. Studies on Internal Physiological and Biochemical Changes of Xantha Mutant in Melon Leaves

lo L.) variety‘ Baishami No.1’.Its contents of photosynthetic pigment and Chl synthesis precursors,chlorophyll fluorescence kinetic parameters and ultra-structure of chloroplasts were studied with mutant parent‘ Baishami No.1’ as contrast.The results showed that photosynthetic pigment content in mutant leaves was significantly lower than that in‘ Baishami No.1’,while the ratio between chlorophyll a and chlorophyll b was significantly higher than in‘ Baishami No.1’.Chlorophyll biosynthesis was hampered at conversion from prophobilinogen(PBG) into uroporphyrinogen (Urogen Ⅲ).Compared with chlorophyll fluorescence kinetic parameters of‘Baishami No.1’,F

,ФPS Ⅱ and ETR were significantly lower,while qN and NPQ were notably higher than that of ‘Baishami No.1’.There were no significant differences in Fm,FV/Fm and qP .Under transmission electron microscopy,9388-1 leaves were found with underdeveloped chloroplast,containing aberrant thylakiods,fewer graum and starch grains than that of‘Baishami No.1’.Grana lamellae were deranged with a line shape and less stacked,but grana lamellae was crowed together in normal one.These results indicated that the biosynthesis chlorophyll was hampered,leading to aberrant chloroplast and the photosynthetic pigments content changed,and then affected their stability.The mutant kept a higher heat dissipation capacity,which could prevent photosynthetic apparatus from damaged by excessive light energy and maintain the normal growth of plant.

Effect of Different Bag Material on Growth,Yield and Quality of Cucumber Cultivated in Bag

Taking cucumber(
Cucumis sativus
L.) cultivar‘ Jinyou 30’as testing material, corn stalk∶cow dung∶slag=3V∶1V∶1V as cultivation medium,taken permeable woven bag, impermeable woven bag,nonwoven and double color film as 4 treatment bag materials,we investigated different bag materials effects on growth,xylem sap,fruit yield and quality of cucumber cultivated in bags in greenhouse. The results showed that after transplanting 40 days and 55 days, the height of cucumber cultivated in impermeable bags were significantly higher than that of other 3 treatments.The fresh weight of above-ground and dry weight of under-ground,and the nutrient content in cucumber xylem sap was also significantly higher than the other 3 treatments.Moreover,the fruit contents of soluble protein and VC were also higher than that of the other treatments.The results indicated that the impermeable woven bag was the optimum bag material for cucumber cultivated in bag.

Effect of Straw Compound Substrate on Tomato Seedling Culture

Taking‘Zhongza 106’tomato(
Lycopersicon esculentum
Mill.)as experiment material,and traditional peat and vermiculite media as contrast,this experiment studied on the physical and chemical properties and effect of tomato seedling culture with different mixing proportion substrates of decomposed straw,rice husk and sand.The results indicated that the mixing proportion substrates of straw could meet the requirement of seedling media and make the quality of tomato seedling better than that of the traditional peat and vermiculite substrates.Among all treatments,T1(100% straw) and T2(straw:rice husk=75V∶25V) compound substrates could culture tomato seedling with the best quality.Its physiological index was notably superior t o the other treatments and contrast.

Counting root tip chromosome numbers,artificial pollination experiments and flow cytometry,etc. were used to confirm that
Taraxacum coreanum
Nakai was tetraploid. Artificial pollination experiments showed that Taraxacum coreanum reproductive type was facultative apomixes, and apomixes rate was around 50%.The result of testing by flow cytometer showed that the ratio of seed cell ploidy level was 2C∶3C∶4C.The reproductive type of tetraploid T. coreanum was facultative apomixes with independent endosperm development.

Status of Pesticide Residue and Regulation Analysis in Vegetables Produced in Inner Mongolia during the Past Decade

Mathematical statistic analysis was carried out on 6 800 samples of veggies,fruits, beans,nuts and seeds produced in Inner Mongolia. The results showed that the dominated vegetables in Inner Mongolia were green leaf vegetables,potato family vegetables and melons,and their yields accounted for 70% of the total vegetables production. Since 2003,with the rapid increase of detected vegetable numbers,the status of vegetable quality safety had entered into a new stage of sustainable steady after a rapid decline or periodic fluctuation,and the differences in safety levels were obvious,of which the range of exceeding standard rate of pesticide residues in eggplants,melons and beans(about 55% of the total vegetables)was under 2%,with a mean of 1.1%,1.6% and 3.1%,respectively. They belonged to stable type. The exceeding standard rate of pesticide residues in green leaf vegetables and Chinese cabbage(about 30% of total vegetables) presented a decreasing trend year by year,wondering in the range of 7%-10%.They belonged to risk main body type.The time period of exceeding standard of pesticide residues in root vegetables and cabbage was under predictable,and in some years its exceeding standard rate was over 5%(amplitude variation was over 15 percentage points).They belonged to random risk type.The kinds of pesticides,which exceeding standard rate in vegetables were relatively intensive,presented variance in different vegetable species.70% of the pesticides belonged to intermittent over-limits and the probability was below 5%.About 20% tradition pesticides often exceed standard and their probabilities were over 30%.The exceeding standard of organophosphorus and carbamates pesticides in vegetables presented a decreasing tend,while the risk of some new type pesticides containing nitrogen atoms and high effective pyrethroid insecticide showed an increasing tendency.

Studies on Effects of Controlling Watermelon Fusarium Wilt by Grafting and Biological Control

Fusarium wilt is an important factor causing clogging in continuous cultivation of watermelon〔
Citrullus lanatus
(Thunb.)Matsum. et Nakai〕.In production,we generally adopt crop rotation,grafting,biological control and medical treatment to prevent this disease.This experiment studied on many controlling methods,such as grafting on different stock varieties,using biological pesticides and biological fertilizer,etc.at different plot of land during the period of watermelon continuous cultivation.The results indicated that the effect of biological control was not obvious,if the continuous cultivation period was long,and watermelon was planted in the fields where Fusarium wilt occurred seriously. Biological control could only play an inhabiting role in dome extent,while the grafted seedling had remarkable effect,the control effect is 100.0%.If the continuous cultivation period was short,and the plot had slightly suffered from Fusarium wilt,it was efficient to apply BIO biological fertilizer or grafting.

Control Effect of Pepper Bacterial Wilt and Stability Test of Mixed Pesticides

Taking 20% Bismerthiazol WP,72% Streptomycin WP,77% Kocide WP a s experiment materials,we found that 20% Bismerthiazol WP mixed with 72% Streptomycin WP could prevent and cure pepper bacterial wilt.The average control effect could reach 96.51% with better stability,which is worthwhile to extend.

A New Potato Variety —‘Longshu No.11’

Longshu No.11’ is a new potato variety selected from L9712-2(92-29-81×93-1-204) as female parent with‘ Yuanza 22’ as male parent.It is of late maturity. Its growing period is about 124 days. Its tuber is round with yellow micro-textured peel,yellow flesh and medium shallow eyes. The average content of dry matter,starch,crude protein,VC and reducing sugar is 23.7%,18.68%, 3.16%,144.2 mg·kg
-1
and 0.162%,respectively.It has higher resistance to late bligh t than‘ Longshu No.5’and has better resistance to leaf roll virus in field.It is sui table for open field cultivation in highaltitude, cold and wet,and semi-arid region. It can yield about 20.25 t·hm-

A New Variety of Yellow Flesh Seedless Watermelon —‘Shenyu’

Shenyu’ is a new watermelon variety with small and medium size fruit,yellow flesh, and seedless.The whole growth period of this variety is about 100 days.It takes about 30 days from female flower bloom to fruit mature.The plant grows in medium and strong vigor.Cauline leaf is in medium level.It has strong resistance to disease and stress. The fruit is of short oval shape with dark green peel.The thickness of fruit peel is about 1.0 cm.The fruit has bright yellow flesh,tastes crispy and refreshing with less fiber. Its seedless performance is good.The central soluble solid content is 11.5%-13.5%.Edge soluble solid content is 7.5%.Its single fruit weight is 2-4 kg.It can normally yield about 27-30 t·hm

A New Sweet and Waxy Corn Variety—‘Gantiannuo No. 3’

‘Gantiannuo No.3’ was a new colored waxy corn bred by crossing double heterozygote (J38×J40)as female parent and sweet waxy inbred line‘Tiannuo 7’as male parent. J38 and J40 were waxy corn with white seed coat and purple seed coat,respectively.The sweet waxy inbred line‘ Tiannuo 7’carries the homozyogous super sweet and waxy genes.‘Gantiannuo No.3’ is a new table corn.Its plant height is 232 cm,with 105 cm ear height. It is resistant to lodging with good stay-green ability and wide adaptation. Its edable quality is superior.The ear shape is mainly of cone type. The fresh ear can yield about 16.5 t·hm
-2
.It takes about 106 days from planting to harvesting.‘Gantiannuo No.3’is suitable to be planted at spring maize area in Northwest China.
