11 Morphological Characteristics of Flower Organ and Cytological Observation of Microspore in Different Development Stage in Radish(Raphanus Sativus L.)

The developmental stage of microspores was observed by two different methods: acetic magenta staining and DAPI staining.It was found that the nucleolus in microspore display more clearly by DAPI staining than acetic magenta staining.The relationship between microspore developmental stage and morphological characteristics of flower buds and anthers was studied by both of cytological and morphological methods on four different radish genotypes.There were significant differences in longitudinal diameter and transverse diameter of flower buds,the ratio between longitudinal diameter and transverse diameter of flower buds,anther length,anther width,petal length,the ratio of petal length and anther length,during the developmental stages of the microspore in all four different radish genotypes.At the late-single nucleus stage,significant difference existed in all the above-mentioned index of different varieties.It indicated that the longitudinal diameter of buds,the ratio of the longitudinal diameter and the transverse diameter of flower buds,petal length and the ratio of the petal length and the anther length were suitable for the determination of sampling time for microspore culture.The appropriate longitudinal diameter of buds ranged 2.991-3.510 mm and the ratio of the longitudinal diameter and the transverse diameter of flower bud ranged 1.412-1.480,the petal length ranged 1.889-2.561 mm,the ratio between petal length and anther length ranged 0.923-1.057.In addition,it is necessary that the anthers should be covered by the sepal,the length of sepal was equal or close to the anther lenghth,and the flower bud was full,the anther showed from light yellow to yellow.

16 The Effect of DNA Demethylation on Vernalization in Broccoli(Brassica oleracea L.var.italica Plenck)

Abstract Taking two varieties of broccoli‘Qingfeng 103’and‘Luxiu’as materials,we adopted different 5azaC treatments on them at young seedling stage,tested the influence of DNA demethylation on vernalization and C/N during vernalization in broccoli.The results showed that the 5-azaC spraying treatment could promote vernalization and shorten flower-bud differentiation stage,and its ratio of total sugar to total nitrogen,soluble sugar to soluble protein,sucrose to soluble protein were higher than the root fertilizing treatment,CK-W and CK-T.

19 Impacts of Plantlet Age and Cultivation Substrates on Domestication Survival Rate of Transgenic Plantlets of Cucumis sativus L.

Taking transgenic plantlets of Cucumis sativus L.as material,we studied the impacts of plantlet age,cultivation substrats and sucrose concentration in rooting media on root development and domestication survival rate of Cucumis sativus L.transgenic in vitro culture plantlets.The results showed that the best root development and the highest survival rate can be obtained when the plantlets were rooted in 1/2 MS media with 6 % sucrose for 28 days and transformed to the same media with 3 % sucrose for 7 days,then transplanted in the cultivation substrates of peat ∶perlite ∶river sand=1 V ∶1 V ∶1 V at 6-true leaf stage.

21 Analysis of Genetic Diversity in Luffa via Morphological and RAPD Markers

In our study,the genetic diversity in 26 luffa accessions were assessed via morphological and RAPD markers,and the genetic relationship among these accessions was revealed by hierarchical cluster analysis.Coefficient of variation based morphological traits ranged from 5.27 % to 107.00 % with the average of 32.75 %.A total of 200 10-mer random primers were used in this study.16 screened primers produced 145 extending and repeatable RAPD bands,of which 125 bands(86.21 %)were polymorphic across 26 luffa accessions.The average Shannon’s information index of RAPD loci was 0.325.The 26 luffa germplasms were clustered into 2 groups based on the morphological markers and RAPD markers by the method of hierarchical cluster analysis respectively.But the clustering results of RAPD markers are not identical with the morphologic classification.

26 Identification Technology for Resistance to Target Leaf Spot of Cucumber and Identification for Variety Resistance

Pycnoconidia solution of 30 000-40 000 per mL was used as identification method for resistance to target leaf spot of cucumber.The solution was sprayed on to the seedlings with 1 true leaf and kept in moisture for 24 hours.Inoculation results showed that 3 accessions were high resistant and 2 accessions were resistant to target leaf spot.‘Jinyou No.3’and‘Jinyou No.38’were high resistant varieties to target leaf spot.

28 Breeding and Utilization of Male Sterile Line 117A in Radish

The original male sterile source from the radish CMS‘Zhe 3A’was transferred to radish inbred line 117.An excellent new male sterile line 117A with good economic traits and stable sterility was obtained by crossing and backcrossing.The rates of male sterile plants of 117A was 100 %,and the traits of flowering and seeding were excellent.The cross combinations using 117A had showed strong heterosis.

31 A New Green Radish F1 Hybrid —‘Weiluobo No.1’

‘Weiluobo No.1’is a new green radish F1 hybrid developed by crossing VL01-17 with VL01-04.Its growth period is about 80 days.It has vigorous growth vigor.The plant height is 35 cm and the plant spread is 48 cm.The plume-like leaf cluster is semi-uprush with small tassel.The leaf color is deep green,but the leafstalk is light green and leave numbers per single plant are 10-12.Its fleshy root is of long column shape with rootlet,little fibre and deep green peel.The length is 26-32 cm,the diameter is 8-12 cm and the weight could reach 2-3 kg.The flesh is light green,with delicate and slight pungent taste.And its dry matter content is 7.0 %,VC 209 mg•kg-1,soluble sugar 3.27 %,coarse fibre 0.65 %.Besides,‘Weiluobo No.1’had high resistance to virus,resistance to downy mildew,stronger resistance to bacterial soft rot than‘Luluobo No.1’.The output can reach 52.5 t•hm-2.So it is suitable to be cultivated in Shandong province and the other areas fit for planting green radish variety.

33 A New Hot Pepper F1 Hybrid —‘Tongyan No.3’

Tongyan No.3’is a new hot pepper F1 hybrid developed by crossing self-line 97-3 selected from local‘Heikezhaojiao’in Nantong with self-line 87-2 selected from the north of China.It is long lantern shaped with length of 10.8 cm,shoulder diameter of 4.8 cm,fruit shape index of 2.3,flesh thickness of 0.23 cm.The average fruit weight is 44.5 g.The yield is 45 t•hm-2.It is suitable to be cultivated both in protected field or after autumn in the middle and lower reaches of Yangtze River.

35 A New Hot Pepper F1 Hybrid —‘Pingjiao No.5’

‘Pingjiao No.5’is a new pepper F1 hybrid developed by crossing self-line 0136(female parent)selected from the local cow-horn shape in Northeast China with selfline 0016(male parent)selected from the sweet pepper in Taiwan.The fruit is of coarse and blunt cowhorn shape.The young fruit is green and yellow and the ripe fruit is bright red,with length of 13.3 cm,shoulder diameter of 5.3 cm,and flesh thickness of 0.35 cm.The average fruit weight is 92 g.It tastes very crisp and tender,with moderate-hot,polished peel.Its yield is about 63 t•hm-2.It has stronger resistance against Phytophthora blight and anthracnose than‘Daniujiao’.It is suitable to be cultivated in both open and protected fields in Northwest China,Henan province and Sichuan province etc.
