1 Research Progress on Regulation and Synthesis Genes on Glucosinolates Biosynthesis in Crucifer

Glucosinolates is a group of important secondary metabolites found in cruciferous plants. They and their degradation products have important biological properties. In recent years, with genome sequencing and functional analysis in model plant Arabidopsis thaliana, people understand more about metabolic pathways of glucosinolates. Its gene regulation and synthesis has become a research hotspot. Some gene families(such as MYB, MAM, CYP79/CYP83, and AOP)have played important roles in different stages of glucosinolate synthesis. The biological functions of glucosinolates and the regulating and synthesizing genes involved are reviewed and the prospect of their application in crucifer breeding is also discussed in this paper.

7 Research Progress on Transgenosis Technology of Chinese Cabbage Group

The essay summarizes the status quo of genetically-modified Chinese cabbage group research in terms of insect-resistance, disease-resistance, stress-resistance, herbicide-resistance, male-sterile, quality improvement and screening marker. It also analyzes the existing problems and points out that the research focus will still remain on establishing regeneration system with high frequency and other effective method. The genetic excursion of transgenic Chinese cabbage group, its influence on other non-target biology and whether it will harm human health will be the hot topics for future research.

26 Construction of RNAi Expression Vector of Pectatelyase from Strawberry

In order to enhance the firmness of strawberry(Fragaria ananassa Duch.)and extend the shelf life, this experiment constructed RNA interference(RNAi)expression vector using a 382 bp conservative fragment of pectate lyase. The target gene was successfully transferred into strawberry via Agrobacterium. Two postitive transformants were detected through PCR analysis.

31 Vanish Dynamics of Chlorpyrifos in Spinach and Effect of Post-harvest Washing on Spinach Safety

The residue vanish dynamics of chlorpyrifos in spring open-field spinach(Spinacia oleracea L.)after sprayed with 750 fold dosage(48 % emulsion), and the effect of post-harvest washing on microbe and chlorpyrifos of spinach were investigated to direct the safety management of spinach from field to market. The results showed that the first class degradation dynamic model of chlorpyrifos in spinach was C=9.542 2e-0.361 t,which predicted the half-life period of chlorpyrifos was 1.92 d, and the safe interval time for spring open-field spinach was 12.6 days when met the standard of GB 2763—2005, which required maximum residue limit of chlorpyrifos was 0.1 mg·kg-1. Postharvest washing for 1 minute by tap water, 5 mg·kg-1 chlorine dioxide(ClO2), 10 mg·kg-1 ClO2, 100 mg·kg-1 hypochlorite(NaClO)and ultrasonic(26 kHz and 450 W)respectively reduced 1 log total bacterial counts, but there still existed 270 cfu·g-1 coliform after treated by 10 mg·kg-1 ClO2, which meant the microbe safety was still a problem, even if some disinfectants were used for commercial washing. NaClO was found to help reducing the residue of chlorpyrifos and ultrasonic was a good washing assistant technology for reducing the residue of chlorpyrifos and microbe counts. The single washing and disinfecting technology could not entirely assure the safety of spinach, so the combined technologies including two or more technologies were suggested to improve the safety of spinach and postharvest fruits and vegetables.

36 Establishment of Vegetable Coconut Coir Cultivation System under Extreme Environment in Torrid Islands Region of China

We studied and established a vegetable coconut coir cultivation system fit for torrid island regions of our country under extreme environment, such as high temperature, high humidity, strong illumination, varied typhoon, water and soil shortage, etc. The whole system consists of three subsystems: environment control system mainly including anti-typhoon, anti-solarization, anti-rainstorm and anti-corrosion greenhouse, fertilizer and water supplying system and coconut coir planting system. It has an obvious effect on anti-typhoon, anti-solarization and anti-rainstorm, but a poor performance on controlling of temperature and humidity. Comparing to vegetable plot in these islands, suitable cultivated vegetable species and their yields are significantly increased. The water saving rate has reached over 70 %.

41 Changes of Several Substances during Ripening and Senescence Process of Cucumber Fruit

In this paper, fruit morphology and several important physiological and biochemical identification indexes correlating with ripening and senescence were monitored for cucumber(Cucumis satious L.)fruits from two different cucumber cultivars DO313 and 649. The results showed that LOX activities were increasing in early stage and decreasing later. The contents of soluble protein in these two varieties had a speedily decreasing process. GS activity was stabilizing in early stage and increasing rapidly in later stage of senescence. The changing trends of these two cucumber varieties were identical, but the existing differences between them may be the causes for their fruit different senescence conducts. The peroxidation of membrane lipid was considered to be one of the major physiological causes leading to senescence of cucumber fruit.

46 RAPD Identification of Black Rind Wax Gourd ‘Heilaocu’ F1 Seed Purity

Using CTAB method, template DNAs were extracted from young roots of germinated wax gourd(Benincasa hispida Cogn.)seeds. Electrophoresis revealed that DNA bands extracted were clear and bright, indicating that the DNA quality could suit RAPD analysis. PCR amplification profiles of black rind wax Gourd variety-‘Heilaocu’ F1 and its parents were analyzed by using 140 RAPD primers. Only one primer, SPS-C15 was found to generate stable and complementary special DNA fragment. A production seed sample was identified with this primer, showing 90.7 % of seed purity. While the seed purity was 89.8 % in field test. The results of these two methods were consistent, suggesting that RAPD technology could be used for rapid testing of seed purity in wax gourd F1 hybrid.

50 Studies on Pre-treatment Method of Determining Mineral Elements in Vegetables by ICP Spectrometer

ICP Spectrometric Method is a high-throughput and accuracy optical test for mineral elements determination. Cucumber, Chinese cabbage, pepper, radish and bean were chosen as materials for mineral elements determination by ICP spectrometric method. Some pre-treatment processes including selection of acid media, drawing method of standard curve, digestion method and different sample digesting state were studied. The results showed that appropriate pre-treatment method can cause some differences on the determination results because of the fresh juicy features of the vegetables. Appropriate pre-treatment method was chosen according to different traits of the samples. Thus, accurate experiment data can be acquired.

55 Inhibitory Effects and Control Efficacy of Paenibacillus polymyxa Strain L1-9 on Alternaria solani of Tomato

L1-9 is a Paenibacillus polymyxa strain isolated from sediment of Lianyungang harbor. In this paper, the antimicrobial activities of L1-9 strain and its fermentations and culture filtrate against Alternaria solani of tomato(Lycopersicon esculentum Mill.)were detected through dual culture methods on PDA medium and agar-well diffusion method. The results indicated that fermentations and culture filtrate of L1-9 strain had obvious antimicrobial effects on restraining conidiophores sprouting of Alternaria solani strain. The inhibition zones got 17.6 mm and 15.8 mm, respectively, after 4 days culture. Fermentations and culture filtrate of L1-9 strain could restrain germ tube elongation of Alternaria solani strain. Moreover, fermentations had stronger antimicrobial effects than its culture filtrate. Biological control functions in laboratory were tested by 2 screening methods: detached leaf method and pot method. The results showed that fermentations of L1-9 and its culture filtrate had better control efficacies against tomato early blight. And the control efficacies of fermentations are better than culture filtrate. The protective and therapeutic effects got 79.28 % and 67.20 %, respectively.

60 Fungicides in Vitro and in Vivo Screening against Fusarium oxysporum f. sp. cucumerinum

Seventeen common fungicides were chosen to screen the effectiveness of cucumber(Cucumis sativus L.)against Fusarium wilt in vitro and in vivo. The results of antibacterial activity assay in vitro showed that the antibacterial activity of hexaconazole, flusilazole, myclobutanil, ziram, thiram, mancozeb, pyrimethanil, thiophanate-methyl and carbendazim was higher, and the inhibition rate in vitro was between 69.61 %-100.00 %. Potted test in vivo showed that the anti-effect of azoxystrobin, kresoxim-methyl, enestroburin, pyrimethanil, carbendazim, thiophanate-methyl was between 61.50 %-95.90 %, which can be used as soil treatment agent for controlling cucumber Fusarium wilt.

66 Screening of RAPD Tagging and SCAR Conversion Related to Aphid Resistant Gene in Pepper

In this study, F2 plants from the cross combination of aphid resistant pepper(Capsicum annuum L.)cultivar 03-C79′, and aphid infection A0003 were used to screen the molecular marker linked to aphid resistant gene in pepper with RAPD molecular marker and BSA(bulked segregation analysis)method. RA750, a specific primer of polymorphism, was obtained from screening 200 RAPD primers for a pepper with aphid resistant traits. The marker from the results of PCR was tested in individual DNA reaction for F2 population and the RA750 amplified fragment could only be detected in the aphid resistant plants. The distance between RA750 and gene was 6.03 cM. According to the sequence of fragment analysis, one pair of primers had been designed. The RAPD marker was converted into a SCAR marker which was named RA750-S.

71 Effect of Adding K,Ca,B,Si on Disease Resistance to Verticillium wilt(Verticillium dahliae)of Eggplant

The effects of four elements, K, Ca, B, Si, on disease resistance, seedling growth and activities of defense enzymes of eggplant were studied by inoculating Verticillium dahliae in field test. The results showed that the disease incidence and disease severity index of eggplant Verticillium wilt were decreased after spraying the solutions of K, Ca, B, Si as compared with the control. And the stem height, stem diameter, root length, fresh matter, root vigor and activities of defense enzymes(POD, PPO and PAL)of eggplant were promoted at low concentrations. While the promotion dropped down with the increase of concentration. Among all the treatments, 50 mg·L-1 of B had the best effect on disease resistance. The disease incidence was lower than that of the control. In this treatment, all growth indices were significantly higher than those of the control. Meanwhile, root vigor and activities of POD, PPO and PAL were 286 %, 20 %, 78 % and 87 % of the control, respectively. It indicated that B could markedly induce the disease resistance of eggplant to Verticillium wilt, and apply to production.

77 Resistance Monitoring of Three Kinds Organophosphorus Pesticides to Prodenia litura(Fabricius)

In order to understand the different resistance level of Prodenia litura living in different regions to the 3 kinds of organophosphorus pesticides used in common, toxicity tests to the 3rd instar larvae of Prodenia litura were carried out in laboratory using leaf dipping method. The result showed that resistance level to Chlorpyrifos increased slowly, the highest level only achieves 5.57 times of the original. Phoxim takes the second place. Its resistance level achieves 2.57-20.74 times. While the highest resistance multiple to Acephate can achieve 109.46 times. The toxicity of these 3 kinds pesticides from high to low was Chlorpyrifos, Phoxim, Acephate.

80 A New Tolerant Storage-transportation Tomato F1 Hybrid —‘Aolin 618’

‘Aolin 618’is a long-shelf life cultivar F1 hybrid released by crossing female parent 61-3-18-15 and male parent 68-5-3-1. This cultivar is outstanding for its early maturity and high potential yield. It is 60-65 days from sowing to harvesting. The first inflorescence appears in the 8th inter-node and the average yield of the 7 inflorescence fruits can reach 105-120 t·hm-2 in open field, and 150 t·hm-2 in greenhouse. Its fruit is red in color with smooth and bright skin. The average fruit weight is about 200 g. The fruit shape index is 0.78. The fruit is 0.9 cm in diameter. The fruit can change color quickly and evenly without green shoulder. This cultivar is highly resistant to late blight and resistant to bacterid wilt. and physiological stress. It is tolerant to low temperature, weak illumination and high humidity. It is also good for storage-transportation. This cultivar is recommended for greenhouse cultivation in early spring and late autumn in northern China; for open field over winter in southern region and over summer at some mountainous areas.

83 A New Variety of Pumpkin —‘Yinhui No.2’

‘Yinhui No.2’was released by systematic breeding involved in segregation populations, which was obtained from crossing European pumpkin‘Mali’inbred lines No. 95-23 with‘Hulinmiangua’inbred lines.‘Yinhui No.2’showed strong growth vigor and stronger resistance. There was fewer branching in its main stem. The first flower grew at the 6-8 axilae, the fruits grew at the 10-12 axilae. Its growing period was about 115 days. Its fruit was oblate. The mature fruits were gray in color. The fruit was 4-5 kg in weight, with 3-5 ventricles. The seed number per fruit was 300-350. The hundred-seed weight was 35-40 g. Its seed was snowy white in color, 1.2 cm in width, and 2.16 cm in length. The seed yield was 1.1-1.5 t·hm-2. It is resistant to virus disease and powdery mildew.

85 A New Potato Variety with High Starch Content —‘Tianshu No.10’

Potato new variety ‘Tianshu No.10’ was developed for high-starch by crossing female parent‘Zhuangshu No.3’with male parent ‘Zhengshu No.1’. It takes about 126 days from sowing to tuber ripening. It is late mature with round tuber. The tuber is light yellow in color, with yellow flesh and shallow eyes. It produces 4.9 tubers per hill. The average tuber size is 132 g. Its market tuber weight percentage can reach 89.8 %. It is suitable to grow in Tianshui, Linxia, Dingxi, Pingliang of Gansu province.

88 A New Variety of Vegetable Jute —‘Funong No.1’

‘Funong No.1’ is a new variety of vegetable jute released by ‘Taizi No.4’ jute after 60Co γ-ray treatment in a dosage of 219 Gy through several generations’selection. Its stem, petiole, stipule, calyces and capsule are all green. The leaves are single alternative and of long-ovate shape. The average length is 16.5 cm, and width is 7.8 cm. The leaf margins serrated and leaf base looks like a pair of teeth extending into tassel shape. After picking asparagus, the leaf height can be controlled around 130-160 cm and its branches numbers are about 15. The stem diameter is 1.60 cm. The growing speed is very slow in seedling stage, while it becomes rapid in middle and late stage. It takes 170-184 days from sowing to seed maturity and the spear leaf yield is 18.0-22.3 t·hm-2. The field cultivation survey indicates that‘Funong No.1’has stronger resistance to spots on jute anthracnose, blight and stem spot diseases than‘Cuilv’,‘Taizi No.4’ and ‘Kuanyechangguo’.
