1 Advances of Research on Cabbage Genetics and Breeding during‘The Eleventh Five-year Plan’in China

The remarkable achievements on genetics and breeding of cabbage(Brassica oleracea L.)in China during‘The Eleventh Five-year Plan’have been made. A number of excellent germplasm resources have been introduced and innovated. The considerable progress has been achieved in disease resistance, stress tolerance and other conventional breeding techniques and biotechnology, such as molecular marker, microspore culture, genetic transformation. 58 new bred varieties with good commodity, strong disease resistance, and tress tolerance were applied and extended in cabbage production. It is summarized the research progress on the aspects of germplasm resources, new varieties of breeding, and biotechnology of cabbage in China. The problems existed and the prospects are also included.

11 Progress in Protected Vegetable Production and Research during‘The Eleventh Five-year Plan’in China

In recent years, great progress has been made in greenhouse vegetable industry in China. The areas of greenhouse agriculture increased by 10 % every year and different types of greenhouse facilities, production modes and technical systems based on characteristics of certain specific districts were developed. This paper analyzed the characteristics of the progress made in greenhouse vegetable industry during the past 5 years in China. We summarized the key research accomplishments and development in greenhouse facility, biological and functional genomics of vegetable science, nursery technology of greenhouse vegetable, high efficient usage of resources, overcoming of soil sickness, mechanism and regulation of stress tolerance, and system establishment for high quality and safety production. We also analyzed the major problems existing in greenhouse vegetable production and discussed about the future trend for industry development and research activities.

24 Field Trial Overview of Genetically Modified Potato in America

A survey was made into the America database on field trials of genetically modified potato(Solanum tuberosum L.)during 1989 to June 2010 in order to reveal the change characteristics and research trend of genetically modified potato in America. Results indicated that 925 field trials, taking 6.1 % of the total field trials numbers on genetically modified potato, were undertaken in 32 states during the last 20 years, dealing with 8 main target characteristics, and 27 research institutions. The accumulated trial areas reached 129 860.69 hm2. In addition,this paper also analyzes the factors effecting changes in field trial of genetically modified potato in America, and carries out discussion on the trend of transgenic potato breeding.

30 Karyotype Analysis and Compaison of Cole Crops

Karyotype analysis and comparison of 7 subspecies of cole crops were conducted by traditional squash method. The results showed that broccoli(Brassica oleralea L.var. italica P.):2n=2x=18=8m+10sm(2SAT), satellites were observed at the 6th pair of chromosomes; cabbage(Brassica oleralea L. var. capitata L.): 2n=2x=18=8m+10sm(2SAT), satellites were observed at the 6th pair of chromosomes; cauliflower(Brassica oleralea L. var. botrytis L.): 2n=2x=18=12m+6sm(2SAT), satellites located in the 7th pair of chromosomes; cabbage mustard(Brassica alboglabra L. H. Bailey): 2n=2x= 18=8m+10sm(2SAT), satellites were observed at the 7th pair of chromosomes; brussels sprouts(Brassica oleralea L. var. germmifera Zenk.): 2n=2x=18=8m+10sm(2SAT)(first time to be reported), satellites were observed at the 7th pair of chromosomes; collard(Brassica oleralea L. var. acephala DC.): 2n=2x= 18=10m+8sm and kohlrabi(Brassica oleralea L. var. caulorapa DC.): 2n=2x=18=8m+10sm, no satellites were observed at these two subspecies. The karyotype type of all subspecies belong to 2A, presenting a ultimately symmetry karyotype. But the evolution extent is inconsistent among different subspecies. In addition,we also discussed about the genetic relationship and genetic diversity of cole crops.

36 Genetic Analysis of Leaf Color Characteristics with Purple Genes on Chinese Cabbage(Chinese Cabbage×Purple Pakchoi)

F1, F2, BC1 and BC2 generations were obtained through crossing, self-crossing and back-crossing by Chinese cabbage〔Brassica campestris L. ssp. pekinensis(Lour)Olsson〕of green leaves with purple pakchoi〔Brassica campestris L. ssp. chinensis(L.)Makino var. communis Tsen et Lee〕of purple leaves in order to study the genetic characteristics of leaf color. The results showed that leaf colors of F1 hybrid were purple, but its leaf colors were lighter than those of purple pakchoi. The ratios were close to 3 purple:1 green for F2 progenies, 1 purple:1 green for BC1 progenies. For BC2 progenies, all plants owned purple colors. The results of χ2 test further proved that heredity of leaf color with purple genes on Chinese cabbage was controlled by one pair of gene. Purple genes were dominant genes compared with green genes and they showed addition effect.

39 Genetic Relationship between Chinese Cabbages in Anti-clubroot with Orange Head Traits

Taking the existing anti-clubroot CR9112A and CR9112B lines of straight canister folded common Chinese cabbage〔Brassica campestris L. ssp. pekinensis(Lour)Olsson〕as female parent and stably inherited‘Longyuanhong No.1’JH-1 inbred lines of Chinese cabbage with orange head as male parent, we prepared every generations of F1, F2 and BC1F1. The inheritance rule of‘Longyuanhong No.1’inbred lines was verified, and the relationship between clubroot resistance gene with orange head leaf gene was explored. The identification results showed that there was no linkage relationship between clubroot resistance and orange head characteristics, and these 2 were independently inherited. The gene type of‘Longyuanhong No.1’JH-1 was msmsrrhh. On this basis, the paper put forward a genetic model for transferring clubroot resistance to orange head traits, so as to obtain an excellent orange head Chinese cabbage resistant to clubroot.

44 Cloning and Expression Analysis of Expansion Cs-EXPA2 Gene in Dwarf Cucumber

In this study, we cloned an expansion gene Cs-EXPA2 from dwarf cucumber(Cucumis sativus L.)D0462 by RT-PCR. We quantitatively detected its expression in hypocotyls, roots and cotyledons within 88 h during cucumber seedling stage by Real-time PCR. The result shows that the level of its expression may be related to the state of cell growth and division in these sites, and the expression level shows obvious differences at different growth stages. The express quantity of Cs-EXPA2 changes along with temporal and spatial variation. The expression pattern of Cs-EXPA2 in hypocotyls is‘low-high-low’, and the expression pattern in roots is‘high-low-high’.

48 Optimization of SRAP-PCR System for Tomato Based on Orthogonal Design

The orthogonal design was used to optimize SRAP-PCR amplification system at 4 levels of 5 factors in tomato(Lycopersion esculentum Mill.)(DNA template, primer, Mg2+, dNTPs and Taq DNA polymerase). The results showed that the order of each factor in different levels affected the result of PCR was: primer>Taq DNA polymerase>dNTPs>DNA template>Mg2+. The most suitable SRAP-PCR reaction system for tomato was total 20 μL containing 15 ng DNA template, 0.75 μmol?L-1 primer, 2.0 mmol?L-1 Mg2+, 0.125 mmol?L-1 dNTPs and 1.0 U Taq DNA polymerase.

53 Identification of Cabbage‘Xiyuan No.4’Seed Purity by SSR Markers

Cabbage(Brassica oleracea L. var. capitata L.)‘Xiyuan No.4’and its parent were analyzed by SSR molecular marker technology. A primer, with obvious different bands parent, O112-G04 was screened from 90 pairs of SSR primers. The DNA fingerprinting of cabbage‘Xiyuan No.4’was constructed, and its seed purity was identified. The result is consistent with those obtained in the field tests. The seed purity was 96.7 %. This result shows that it is reliable to use this pair of SSR primer to identify cabbage‘Xiyuan No.4’seed purity.

56 Studies on Heat Tolerance Identification Method for Non-heading Chinese Cabbage〔Brassica campestris L. ssp. Chinensis(L.)Makino var. communis Tsen et Lee〕

Ten non-heading Chinese cabbages〔Brassica campestris L. ssp. chinensis(L.)Makino var. communis Tsen et Lee〕materials were continuously treated with 6 different high temperatures for 7 days in artificial climate box in spring, autumn and summer, in order to identified their heat tolerance. The results indicated that under different heat stress, different degrees heat injure symptoms such as leaf back-scrolling, chlorosis, appeared in all tested materials,and these symptoms became severe along with temperature increasing and treatment time prolonging. The heat injure symptoms could be intensified. The heat injury index at seedling stage by heat treatment of 37 ℃/27 ℃(d/n)for 4 days in spring and autumn could significantly reflect the differences in heat tolerance of different non-heading Chinese cabbage varieties. While in summer, culturing seedling with nature temperature under 37 ℃/27 ℃(d/n)could not significantly distinguish the differences in heat tolerance of different non-heading Chinese cabbage varieties.

62 Effects of Grafting on Rhizosphere Microorganisms Quantity and Community Structure of Watermelon

The changes of microorganisms population, bacterial community structure and 2, 4-DAPG producers community structure in rhizosphere soil of grafted watermelon〔Citrullus lanatus(Thunb.)Matsum. et Nakai〕plants were studied using the methods of microorganism culture, 16S rDNA PCR-RFLP and phl D gene PCR-RFLP, respectively. The results showed that the bacterial and fungal population in rhizosphere soil of grafted watermelon was increased and that of the actinomyces population was decreased; the grafted watermelon had one similar main 16S rDNA genotype in rhizosphere soil with the non-grafted watermelon, although the grafted watermelon with different stock had their unique 16S rDNA genotypes different from non-grafted watermelon; the grafted watermelon had different main 2, 4-DAPG producers phl D genotypes from non-grafted watermelon.

67 Effects of Iodine on Fruit Quality of Cherry Tomato in Nutrient Solution Culture

In controlled condition of deep flow hydroponics, the experiment studied the effect of iodine on fruit quality of cherry tomato(Lycopersicon esculentum var. cerasiforme)treated in solutions with iodide(I-)concentrations of 1.0-6.0 mg?L-1. The results showed that there were some influences on the iodine, VC, total acid, soluble sugar and moisture contents of cherry tomato fruit. The iodine content in fruit increased with the increasing of I- concentration,and each treatment was significantly higher than that of the CK. The correlation between iodine in fruits and I- concentration in solutions reached extremely significant level, with the correlation coefficient of 0.947. The VC content in the fruit, of which the plants were treated with I- concentration of 3.0 mg?L-1 was the highest, being 9.7 % higher than the CK or 201.7 mg?kg-1 of fresh weight. There was no significant difference between the total acid in fruit with low I- concentration(1.0-3.0 mg?L-1)treatment and the CK, the minimum of total acid in fruit being 0.64 % with I- concentration of 3.0 mg?L-1; but for plants treated with high I- concentration(4.0-6.0 mg?L-1), it was significantly higher than the CK. The soluble sugar in fruit significantly reduced, when the plants were treated with I- solution, but there were no significant differences among the soluble sugar in fruit, when the plants were treated with I- concentration from 1.0 mg?L-1 to 5.0 mg?L-1. The fruit sugar acid ratio had no significant difference between the plants treated with I- concentration of 3.0 mg?L-1 and the CK. When the plants were treated with high I- concentration(4.0-6.0 mg?L-1), the fruit moisture content was significantly lower than the CK and 1.0-3.0 mg?L-1 I- concentrations, but there was no significant difference between the plants treated with low I- concentration(1.0-3.0 mg?L-1)and the CK. To summarize these results, it can be concluded that when the I- concentration of 3.0 mg?L-1 was provided for the growth of cherry tomato, fruits with high vitamin Ccontent, low total acid and moderate iodine content will be obtained, though the influence of the I- concentration on the sugar acid ratio and moisture content were not significant.

73 Measuring Plant Leaf Area by Scanner and ImageJ Software

The accuracy of measuring tomato(Lycopersicon esculentum Mill.)leaf area by ImageJ software was confirmed through measuring scanned standard shape. There was no significant difference for area with 100, 200 and 300 DPI. And 200 DPI was better. Two methods of paper-cutting and image processing for measuring tomato leaf area were compared and there was no significant difference. Based on the result, the method of measuring tomato leaf area with scanner and ImageJ software was established.

78 Studies on Cultivation Pattern of Straw Wrapping Potato in Winter- planting

To solve the existing problem in straw no-tillage potato(Solanum tuberosum L.), the comparative study on 3 cultivation patterns were performed. The results showed that all these 3 cultivation patterns were better than the conventional pattern. Among them the straw wrapping and coverage with black film in winter-planting potato could shorten the germination time, increase the commercial potato rate and potato numbers of single plant, improve the potato quality of single plant, remarkably raise the yield and efficiency. The straw wrapping and coverage with black film all precede the straw no-tillage pattern. The net income from straw wrapping pattern can reach 6 042.15 Yuan per hm2 and the ratio between investment and output is 1.76.

81 Effect of Sodium Alginate on Raphanus sativus L. Seedlings in Adaptation to Salt-tolerance

The effect of sodium alginate on Raphanus sativus L. seedlings in adaptation to salt- tolerance was studied by water culture. The results showed that after culture solution seedlings of Raphanus sativus L. were dealt with NaCl concentration at 3-12 g?L-1, adding sodium alginate concentration at 0.5 g?L-1, the seedlings survival rate was increased by 41.1 %, the number of root was increased by 34.9 %, the single plant seedlings weight was increased by 34.7 %, and the SOD activity was increased by 35.4 %. All these indicated that sodium alginate can increase salt-tolerance adaptability of Raphanus sativus L. seedlings, and relief the harm brought about by NaCl stress.

85 Determination of Trapping Efficiency of Traps with Sex Pheromone of Prodenia litura and Analysis of Its Affecting Factors

With mark-release-recapture methods, the trapping efficiency of traps with sex pheromone of Prodenia litura and the effect of meteorological factor on trapping efficiency were studied in the field. The results showed that the trapping efficiency was from 7.46 % to 72.46 %. The effective trapping radius of were from 20 m to 40 m. The trapping efficiency to Prodenia litura was determined by meteorological factors: like wind direction,wind speed and the relative humidity. The largest number of Prodenia litura attracted by traps was in the windward of dominant wind direction. Higher speed of wind and lower relative humidity could promote the trapping efficiency at night.

89 Effect of Ultrasonic Washing Technology on Cleanliness of Carrot and Pakchoi

Taking carrot(Daucus carota L. var. sativa DC.)and pakchoi〔Brassica campestris L. ssp. chinensis(L.)Makino var. communis Tsen et Lee〕as experiment materials, we studied the cleanliness of ultrasonic washing technology, which involved different frequencies, powers and washing time. The results showed that cleanliness of carrot and pakchoi can be improved effectively by treatment of 26 kHz, 0.32 W?cm-2 for 10 min and 26 kHz, 0.28 W?cm-2 for 2 min, respectively. Both of these 2 treatments can meet the requirements for commercial vegetable appearance.

92 A New Chinese Cabage F1 Hybrid —‘Jingcui No.60’

‘Jingcui No.60’is a high quality Chinese cabbage F1 hybrid bred with microspore culture technology. It is an early-maturity variety. It can be harvested in 60-65 days after sowing. The plant type is up righting and the plant height is 58 cm. Its leaf is of long downside egg shape. Its leaf is of dark green color with many wrinkles. Its leaf head is of long tube shape with green color. The leaf head is 40 cm in height, 13 cm in diameter. The leaf head weight is 2.0-2.4 kg. This variety is of excellent quality, high resistant to virus disease, resistant to downy mildew and black rot. It can yield about 67.5 t?hm-2. It is suitable to grow in Beijing, Hebei, Inner Mongolia, Liaoning, Jilin, Yunnan, Guizhou and Guangxi. The total extension area is nearly 1 000 hm2.

94 A New Cabbage Variety —‘Xiyuan No.11’

‘Xiyuan No.11’is a new cabbage F1 hybrid developed by crossing self-incompatible line 2004G with self-incompatible line 2004I. Its growing period is 165-185 days. The head is flat round and compact with tender leaves and good quality. Its VC content is 344 mg?kg-1(FW),fiber content is 5.8 g?kg-1(FW)and soluble solid content is 5.1 %. The average single head weight is about 1.5 kg. It is tolerant to inmanturity bolting. It can yield about 52.5 t?hm-2. It is suitable to be cultivated in Chongqing and Sichuan Provinces.

97 A New Hot Pepper F1 Hybrid —‘Yujiao 17’

‘Yujiao 17’is a new hot pepper F1 hybrid bred by crossing inbred line 101-1(as female parent)with inbred line 104-1(as male parent). Its fruit is of sheep-horn shape, 15.2 cm in length, 3.1 cm in diameter. The average single fruit weight is 50.2 g. The fruit color is yellow-green. It can yield 45 t?hm-2. The VC content is 873 mg?kg-1 and the soluble sugar content is 2.33 %. It is high resistant to virus disease, phytophthora blight and anthracnose of pepper. It is suitable for cultivation both in open and protected fields in early spring.

100 A New Potato Variety —‘Longshu No.9’

‘Longshu No.9’is bred through systematic and oriented selection from the progeny of a novel clone 93-10-237 as female parent and a line Datong G-13-1 as male parent. The variety is with middle-late maturity and about 118 days growth period. Its tuber is of flat round shape, with pale yellow flesh and light yellow and russet skin. The big and middle size tubers take above 95 %. The average dry matter content is 26.18 %, starch is 20.39 %, crude protein is 2.840 %, the VC content is 131.8 mg?kg-1 and reducing sugar is 0.201 %. Its yield is approximately 29.16 t?hm-2. It is suitable to be popularized and planted in areas with high latitude, cold and wet climate, and semi-arid areas.

103 A New Autumn Radish F1 Hybrid —‘Jinghong No.3’

‘Jinghong No.3’is a new autumn radish F1 hybrid bred by crossing male sterile line R01A with male fertile line R5. The root is of round shape with 15.0 cm in length and 13.5 cm in diameter. The root skin is red in color and the internal flesh is crystal white with good edible flavor. The plants are outspreading phyllome with few leaves and lobed leaflets. It takes 70-80 days from sowing to maturity. It has higher resistant to virus and soft rot than‘Hongfeng No.1’, and suitable for autumn growing. The glucosinolates content in root is 480.9 μmol?kg-1(FW), which is nearly two fold higher than that of the control. Its yield in open field is about 75 t?hm-2. It has been extended to Beijing, Hebei and Liaoning Provinces for 66.7 hm2.

105 A New Kidney Bean Variety —‘Liyun No.1’

‘Liyun No.1’is a hybrid kidney bean variety. Its female parent is superior local variety‘Honghuaheizi’and male parent is‘Heizhenzhu’. It has strong growth vigor. The pod length is about 17.2 cm. The single pod weight is about 10.8 g. The pod color is green and the pod can not be easily fibering. It can yield about 24.99 t?hm-2 in open field at mountainous areas, where the altitude is between 550 m to 800 m. It is tolerant to high temperature and drought. It is suitable to be planted in mountainous areas.

107 A Starch-based Water Chestnut Variety —‘Guifenti No.1’

‘Guifenti No.1’is a new starch-based water chestnut variety bred by individual selection and biological technology-cultivated with virus-free tip tissue biotechnology. It is the first variety in China used specially for processing water chestnut powder and approved by provincial authority. This new variety is fast-growing with strong tillering ability, big and uniform corms. There are 90 % corms with horizontal diameter >3.00 cm, 100 % vertical diameter >1.90 cm among 20 corms by random selection. While in the control group, the ratios are only 40 % and 45 %, respectively. Its yield is 28.5 t?hm-2, 5.2 %-27.0 % higher than the control. Its starch content is 12.7 %.

110 A New Tomato F1 Hybrid —‘Dongnong 715’

‘Dongnong 715’is a new mid- maturing tomato F1 hybrid,developed by crossing 675 as male parent and 642 as female parent. It is high resistant to ToMV, leaf mold, Fusarium wilt and Verticillium wilt. The fruit is of large size. The average head weight is 230-250 g. The mature fruit is pink in color with regular shape. The average firmness is 0.61 kg?cm-2. It has good commodity character for storage and transportation. The average yield is 150-225 t?hm-2 in protected fields. It is tolerant to low temperature and has no abnormal fruit under low temperature environment. It is suitable for cultivation in protected fields in China.
